Megastigmatrienone is a family of related norisoprenoid ketones best known as the character impact compounds of cured tobacco aroma.
The molecules share a C13 megastigmane skeleton with a 4,5,7-trien-3-one or 3,5,7-trien-2-one substitution pattern. Commercial material is usually a mixture of four positional and geometric isomers, with formula C₁₃H₁₈O and molar mass 190.28 g/mol. The product is a pale yellow to amber viscous liquid.

The U.S. Food and Drug Administration recognises it as a synthetic flavouring substance under 21 CFR 172.515. JECFA has evaluated the broader norisoprenoid ketone group and placed the compound in a class of no safety concern at current intake levels.
The IFRA does not impose specific restrictions on use in fragrance applications at standard dosing levels.
Manufacturing Process
Megastigmatrienone is produced by chemical synthesis from C13 norisoprenoid precursors.
The classical route starts from beta-damascone or beta-ionone derivatives. Controlled oxidation and isomerisation build the trienone system across the six-membered ring and side chain. The reaction conditions are tuned to give a defined ratio of the four main isomers, since each contributes a slightly different facet to the overall aroma.
An alternative route uses degradation chemistry on carotenoid feedstocks. Selective oxidation of beta-carotene or related precursors generates a mixture of norisoprenoid fragments, from which the megastigmatrienone family can be isolated and enriched by distillation.
The natural occurrence in cured tobacco is the result of enzymatic and oxidative degradation of leaf carotenoids during the curing and aging process, which is one reason the molecules are so closely tied to tobacco character.
