Manufacturing Process

Industrial production of 2-Acetyl Pyridine relies on synthetic routes that install the acetyl group on a pyridine ring.

The most common pilot and plant route starts from 2-cyanopyridine. Reaction with methylmagnesium halide or methyllithium followed by aqueous hydrolysis of the resulting ketimine gives the target ketone in good yield.

An alternative is the directed metallation of pyridine at the 2-position, followed by quenching with acetic anhydride or N-methoxy-N-methylacetamide (Weinreb amide) to deliver the acetyl group cleanly.

Crude reaction product is washed, dried, and purified by fractional vacuum distillation. The finished liquid is filtered and packed under nitrogen to limit oxidative darkening during storage.
