2-Acetyl Thiazole is a pale yellow to amber liquid aroma chemical with the formula C₅H₅NOS and CAS number 24295-03-2. It delivers a warm popcorn note coupled with a distinctive roasted cereal and faintly bready character.

The compound is a member of the sulphur-nitrogen heterocyclic ketone family that defines much of the Maillard top in flavour work. It is dosed at parts-per-billion to low parts-per-million levels in finished food.

Manufacturing Process

2-Acetyl Thiazole is made by synthesis on a pre-formed thiazole ring.

The most widely used route starts from 2-bromothiazole or 2-iodothiazole. Halogen-metal exchange with butyllithium at low temperature, followed by quenching with N-methoxy-N-methylacetamide (Weinreb amide) or acetic anhydride, installs the acetyl group cleanly at the 2-position.

An older route uses Friedel-Crafts acylation of thiazole with acetyl chloride and a Lewis acid catalyst, though regioselectivity is lower and downstream purification is heavier.

Crude product is purified by vacuum distillation under nitrogen. The finished liquid is filtered and packed under inert gas in light-shielded containers, since prolonged exposure to air and light can deepen the colour.
