Aloe Vera Gel
Aloe vera gel is the clear mucilaginous parenchyma from the centre of the leaf of Aloe barbadensis Miller, separated from the green rind and from the bitter yellow latex that lies immediately beneath it. It is traded as single strength liquid, as concentrate at 10, 40 or 200 fold, and as spray-dried powder, and it is distinguished from whole leaf material by the fact that the aloin-bearing latex is excluded mechanically rather than removed later by adsorption.
The gel is more than 98 percent water. The functional fraction is acetylated polysaccharide, principally acemannan, a beta-1,4 linked acetylated mannan whose molecular weight and acetylation degree determine both the viscosity and the biological activity attributed to the material. Organic acids, minerals and small amounts of amino acids and sterols make up the remainder.
The polysaccharide is the fragile part. It is depolymerised by the plant's own enzymes within hours of the leaf being cut, and by heat, prolonged storage and low pH, so a gel with a high acemannan content is the product of fast, cold processing rather than of any additive. Solutions are slightly viscous, near neutral, and clear to slightly opalescent.

Common Markets Grades
Liquid grades are quoted by concentration factor and by total solids, with single strength gel at around 0.5 percent solids, and 10 fold, 40 fold and 200 fold concentrates traded for reformulation. Spray-dried powder is quoted at 100 fold or 200 fold equivalence, usually with maltodextrin as a carrier, and carrier-free freeze-dried gel is a separate and more expensive article.
The meaningful specification beyond solids is acemannan or total polysaccharide content, measured by an appropriate method, since a lot with correct solids but degraded polysaccharide is of little use. Aloin content is specified and held low, normally below 10 mg/kg for the decolourised trade standard, and this is straightforward for inner leaf gel because the latex was never included. Certification under the International Aloe Science Council programme is commonly required as third party assurance of both identity and dilution, since adulteration with maltodextrin and with diluted material is documented in this trade. Preserved and unpreserved liquids are both traded, and organic certified lots are widely available.

Regulatory Status
In the United States aloe vera gel is used in foods, beverages, supplements and cosmetics. Aloe vera gel from the inner leaf has a long history of use in beverages and is treated as a food ingredient, and aloe vera is also listed among the natural flavouring substances. The regulatory line that matters runs between the gel and the latex: the FDA removed aloe latex from over-the-counter laxative products for want of submitted safety data, and aloin-bearing preparations are treated quite differently from decolourised inner leaf gel. Cosmetic use falls under the general safety and labelling provisions.
In the European Union aloe vera gel is used as a food and cosmetic ingredient and carries no E number. Preparations from Aloe species containing hydroxyanthracene derivatives, including aloin and aloe-emodin, have been prohibited for use in food following a safety assessment. The distinction between decolourised inner leaf gel and latex-bearing preparations is therefore a legal one and not merely a quality preference, and suppliers must demonstrate that hydroxyanthracene derivatives are absent. Cosmetic use is governed by the cosmetic products regulation. Codex has not adopted a standard for aloe gel; the general provisions on contaminants and hygiene apply.
Manufacturing Process
Mature leaves are cut by hand and processed within hours, because the enzymes released at the cut surface begin degrading the polysaccharide immediately. Leaves are washed and sanitised, and the butt end is stood to drain so that the yellow latex runs out; this drainage step, together with rind removal, is what keeps aloin out of the inner leaf product. The leaf is then filleted, either by hand with a knife or on a mechanical filleting line, to separate the clear parenchyma from the green rind.
The fillet is macerated or ground and depulped through a screen, giving a viscous juice. It is treated with a cellulase or pectinase where a clear, low-viscosity product is wanted, then filtered, and pasteurised by a short high-temperature hold or by flash pasteurisation. The shortness of the hold is the point, since a long thermal hold destroys the acemannan the gel is bought for. Concentration is by low-temperature vacuum evaporation or by reverse osmosis and ultrafiltration. The membrane routes are preferred because they avoid heat. Liquid grades are preserved and filled; powder grades are spray dried with maltodextrin at a controlled outlet temperature or freeze dried for a carrier-free article, then sieved and packed.
