Kidney protein peptide, lyophilised, is a freeze dried powder of short peptides produced by enzymatic hydrolysis of animal kidney protein, generally porcine or bovine kidney recovered as an edible offal at slaughter.

Kidney tissue is protein dense and, unlike hide or bone, its protein is largely soluble cytoplasmic and membrane associated material rather than structural collagen. Hydrolysing it gives a peptide mixture with a broad amino acid distribution and none of the glycine and hydroxyproline signature that marks a collagen peptide, along with a naturally high content of nucleotides and bound trace elements. Freeze drying is chosen over spray drying to keep those heat sensitive minor components intact, which is the basis on which the material is sold as an active peptide rather than a plain hydrolysate.

The powder is light yellow to pale brown, porous and low in bulk density, with a savoury and distinctly offal like odour that limits its use in unflavoured formats. It dissolves rapidly and completely in water because lyophilisation leaves an open cake, and it is hygroscopic enough to require nitrogen flushed foil packaging and handling in a humidity controlled room.

Common Markets Grades

Species and tissue traceability form the first grade distinction, with porcine kidney the volume feedstock and bovine kidney used where a non porcine source is specified. The second is molecular weight, with general grades holding the greater part of peptide mass below 3000 Daltons and oligopeptide grades ultrafiltered below 1000 Daltons for faster dissolution and a cleaner taste. Protein content on a dry basis usually starts around 70 percent and rises above 80 percent for ultrafiltered material.

Regulatory Status

In the United States hydrolysates of edible offal are used as dietary ingredients in supplements under the Dietary Supplement Health and Education Act, with the manufacturer holding the safety basis, and separately as components of microbiological media and animal feed under their own frameworks. There is no food additive regulation and no published generally recognised as safe listing naming kidney peptide, so conventional food use would rest on a self determined generally recognised as safe conclusion for the specific source and process. Bovine material is subject to the ruminant tissue controls imposed against transmissible spongiform encephalopathy, and kidney is one of the tissues most closely monitored in the national residue programme, so imported material is screened accordingly.

In the European Union a peptide preparation from mammalian kidney has no established history of significant consumption in that form and would normally require novel food authorisation before being marketed as a food or food supplement ingredient. Production is also governed by the animal by-products framework, which determines which categories of material may be processed for food and feed and requires approved establishments, and by the transmissible spongiform encephalopathy rules where bovine tissue is involved. No E number applies, since the material is not an additive, and neither Codex nor JECFA has established a specification for it.

Manufacturing Process

Chilled or frozen kidney from an approved abattoir is trimmed of the capsule, fat and ureteric tissue, split and washed repeatedly to purge residual urine and blood, since inadequate washing carries a strong odour through the whole process. The cleaned tissue is minced with water into a pumpable slurry and heated briefly to inactivate the very active native proteases and to reduce the microbial load, then cooled to the working temperature of the enzyme system.

Hydrolysis is run in a stirred reactor at controlled pH, typically with a bacterial endoprotease to cleave the chains followed by an exopeptidase preparation to trim the hydrophobic terminal residues that carry bitterness, and is tracked to a target degree of hydrolysis before the reactor is heated to denature the enzymes. The digest is centrifuged, which removes undigested tissue and separates the fat layer, and the clarified liquor is ultrafiltered at a few kiloDaltons to hold back residual whole protein. Activated carbon treatment for colour and odour is applied where the grade calls for it. The permeate is concentrated at low temperature under vacuum, filled into trays, frozen solid, and dried in a lyophiliser where the ice sublimes under vacuum without the product melting. The dried cake is milled, sieved, and packed at once under nitrogen.
