Vitamin D2 is the plant and fungal form of the fat-soluble vitamin D family, used in supplement, pharmaceutical, food, and feed formulations as ergocalciferol.
The molecule carries the formula C₂₈H₄₄O, a molar mass of 396.65 g/mol, and the CAS number 50-14-6. Like vitamin D3 it is a secosteroid, generated by ultraviolet irradiation of a sterol precursor, in this case ergosterol from yeast cell walls or from mushroom tissue.

Oil and dry formats of ergocalciferol cover a range of activity-keyed potencies.
Crystalline ergocalciferol at 40 million IU per gram is the reference active substance for pharmaceutical and high-potency supplement work. It is markedly oxygen and light sensitive and is normally handled under nitrogen blanket in amber containers.
Oil-resin grades at 1.0 to 4.0 million IU per gram, diluted in refined corn or medium-chain triglyceride oil, serve liquid drop, softgel, and oil-based pharmaceutical preparations.
Beadlet and cold-water-dispersible grades at 100 and 500 thousand IU per gram, built on modified starch or acacia gum matrices with antioxidant systems, are the standard supply for vegan-certified tablets, capsules, and beverage fortification. Pharmacopoeial grades meet USP and Ph. Eur. monographs.

Regulatory Status
Ergocalciferol is authorised as a nutrient and as a pharmaceutical active substance in essentially every regulated market.
The European Union lists it as a permitted vitamin source under Regulation (EC) No 1925/2006 and Directive 2002/46/EC. UV-treated mushrooms and UV-treated yeast carry separate novel-food authorisations for vitamin D2 fortification of bread, baking, and supplement matrices. EFSA has set a tolerable upper intake level of 100 µg per day for adults, shared with vitamin D3.
The U.S. Food and Drug Administration recognises ergocalciferol as GRAS for specific food categories and approves it as a prescription drug at high dose for the treatment of vitamin D deficiency. The Daily Value sits at 20 µg.
Codex Alimentarius includes it in the Advisory List of Nutrient Compounds. JECFA has reviewed both D2 and D3 and treats them as a group for safety assessment.
Pharmacopoeial monographs appear in the USP, Ph. Eur., JP, and BP. Vegan certification is straightforward because the upstream feedstock is yeast or mushroom rather than lanolin, and Halal and Kosher certifications are also routinely offered.
Manufacturing Process
Industrial ergocalciferol is produced from ergosterol, the principal sterol of fungal and yeast cell membranes.
The starting material is recovered from spent Saccharomyces cerevisiae biomass after brewery, baking, or dedicated fermentation operations. The yeast is autolysed, the lipid fraction is extracted with hydrocarbon solvent, and ergosterol is crystallised from the lipid concentrate to deliver high-purity provitamin material.
Photolysis is the defining step. A thin film of ergosterol in an inert solvent is irradiated with ultraviolet light at approximately 295 nm, opening the steroid B-ring to give previtamin D2. A thermal rearrangement at controlled temperature isomerises the previtamin to ergocalciferol.
The crude photoproduct is purified by chromatography and recrystallisation to deliver high-purity crystal, then dissolved in oil or formulated into beadlets for downstream supply. Production is concentrated in Europe and China. UV-treated mushrooms represent a parallel and smaller route in which whole-food matrices are exposed to UV light during post-harvest processing to generate vitamin D2 in situ.
