Vitamin D3 is the animal-derived form of the fat-soluble vitamin D family, used in supplement, food, pharmaceutical, and feed formulations as cholecalciferol.

The molecule carries the formula C₂₇H₄₄O, a molar mass of 384.64 g/mol, and the CAS number 67-97-0. Activity is expressed in micrograms or international units, with 1 IU equal to 0.025 µg of cholecalciferol.

Cholecalciferol divides into oil and dry formats, with potency keyed to formulation needs.

Crystalline cholecalciferol at 40 million IU per gram is the reference active substance for pharmaceutical and high-potency supplement work. It is highly oxygen-sensitive and is normally handled under nitrogen blanket.

Oil-resin grades at 1.0 to 4.0 million IU per gram, typically diluted in medium-chain triglyceride or refined corn oil, serve liquid drop, softgel, and oil-based pharmaceutical preparations.

Beadlet and cold-water-dispersible grades at 100, 200, 400, and 500 thousand IU per gram, built on gelatin or modified starch matrices with antioxidant systems, are the standard supply for tablets, sachets, beverage fortification, and animal-feed premixes. Pharmacopoeial grades meet USP and Ph. Eur. monographs.

Regulatory Status

Cholecalciferol is authorised as a nutrient and as a pharmaceutical active substance in every regulated market.

The European Union lists it as a permitted vitamin source under Regulation (EC) No 1925/2006 and Directive 2002/46/EC. EFSA has set a tolerable upper intake level of 100 µg per day for adults.

The U.S. Food and Drug Administration recognises cholecalciferol as GRAS under 21 CFR 184.1950 for specific food categories, with broader use permitted through food-additive petitions. The Daily Value sits at 20 µg.

Codex Alimentarius includes it in the Advisory List of Nutrient Compounds. JECFA has reviewed the substance for use in infant formula and follow-on formula.

Monographs appear in the USP, Ph. Eur., JP, and BP. Halal and Kosher certifications are routinely available; vegan certification requires a non-lanolin source.

Manufacturing Process

Industrial cholecalciferol is produced from 7-dehydrocholesterol, the same provitamin found in mammalian skin.

The starting material is isolated from lanolin, the waxy secretion of sheep wool, which is degreased, saponified, and subjected to multiple crystallisations to recover cholesterol. The cholesterol is then converted to 7-dehydrocholesterol via bromination and dehydrohalogenation.

Photolysis is the defining step. A thin film of 7-dehydrocholesterol in an inert solvent is irradiated with ultraviolet light at around 295 nm, opening the steroid B-ring to give previtamin D3. A thermal rearrangement at controlled temperature isomerises the previtamin to cholecalciferol.

The crude photoproduct is purified by chromatography and recrystallisation to deliver high-purity crystal, which is then dissolved in oil or formulated into beadlets for downstream supply. The principal manufacturers operate in Europe and China, with vegan alternatives derived from lichen biomass occupying a small but growing share.
