Ganoderma lucidum spore is the reproductive spore of the reishi mushroom, Ganoderma lucidum, released in vast numbers from the underside of the mature cap and collected as a fine powder that is traded as a nutraceutical raw material in its own right.

Each spore is an oval body of roughly five to eight micrometres with a distinctive double layered sporoderm built largely of chitin. It holds a concentrated version of the constituents found in the fruiting body: triterpenoid ganoderic and lucidenic acids, beta-glucan and heteropolysaccharides, ergosterol and other sterols, spore lipids rich in unsaturated fatty acids, protein, nucleosides and trace selenium and germanium taken up from the substrate.

Collected spore powder is a fine, dense, deep purplish brown to reddish brown material with a faint mushroom odour and a bitter taste. In the intact state it is close to insoluble and largely resistant to digestion, because the sporoderm survives the gut, which is why the untreated powder is traded chiefly as the feedstock for wall breaking and for oil extraction rather than as a finished ingredient. It is hydrophobic and floats on water. Once the wall is broken the exposed lipid fraction oxidises, so even the intact powder is dried carefully, kept from heat and light and packed under nitrogen.

Intact spore powder is graded on purity and on collection method before anything else. Lots are specified on the proportion of foreign matter, since spores collected without proper enclosure carry substrate dust, mycelial fragments and insect debris, and on moisture, which decides both storage stability and microbiological risk. Beyond that the standardised markers are total triterpenes, usually expressed as ganoderic acid, and crude polysaccharide measured as beta-glucan.

The intact powder is normally an intermediate rather than the finished article, since digestibility depends on breaking the wall, and most commercial material is sold on after mechanical disruption or after supercritical extraction of the spore oil. Selenium enriched spore powder, produced by cultivating on a selenium supplemented substrate, is traded as a distinct specification, as is log grown material where the substrate is stated on the certificate. Organic certified lots are handled separately, with substrate composition and heavy metal results the deciding factors because the mushroom concentrates minerals from what it grows on.

Regulatory Status

In the United States, Ganoderma lucidum spore powder is marketed as a dietary ingredient under the Dietary Supplement Health and Education Act, reishi having an established history of use in supplement form. It is not an approved food additive for general food use, so finished products are positioned as supplements rather than as conventional foods, and claims are limited to structure function statements.

In the European Union the position is more restrictive. Preparations from Ganoderma lucidum that lack a demonstrated history of significant consumption in the Union before May 1997 fall under the novel food regulation and require authorisation before being placed on the market, and a spore powder is assessed on its specific production route and specification. Member state rules on botanicals in food supplements apply in parallel. The material is not a food additive, carries no E number, and JECFA has issued no specification for it. Contaminant testing is the practical control, with heavy metals and radionuclides the usual points of scrutiny because the fungus concentrates minerals from its growing substrate, so substrate documentation normally accompanies commercial lots.

Manufacturing Process

Spores are collected rather than harvested. Reishi is grown either on hardwood logs buried in shaded beds or on sterilised sawdust substrate blocks under controlled temperature and humidity, and once the cap edge matures the fruiting body begins releasing spores continuously over a period of days to weeks. Growers enclose the growing beds with fine mesh tents or fit collection hoods and negative pressure ducting that draws the airborne spores into collection chambers and cyclone separators, and the timing matters, since spores released before maturity are lower in the active fractions and material left in the bed picks up contamination.

Collected raw powder is screened over fine sieves and passed through air classifiers and magnets to remove substrate fragments, mycelium, cap debris and insect matter, then dried in low temperature air or vacuum dryers to a moisture low enough for storage stability, since the material arrives damp from a humid growing environment. It is given a sterilisation step, most often short low temperature steam or irradiation where the destination market permits, to reduce the microbiological load carried from the growing house. The dried, cleaned powder is assayed for triterpenes and polysaccharide, blended across collections to a uniform lot, and packed under nitrogen in light barrier drums. From there it goes either to chilled mechanical wall breaking or to supercritical carbon dioxide extraction of the spore oil.
