Hydroxytyrosol

Hydroxytyrosol (CAS 10597-60-1) is the principal phenolic alcohol of the olive tree (Olea europaea) and the polyphenol most closely identified with the cardiovascular profile of olive oil.

The compound has the formula C₈H₁₀O₃ and a molar mass of 154.16 g/mol. It is a colourless to pale yellow viscous liquid or low-melting solid, very soluble in water, ethanol, and most polar organic solvents.

Common Markets Grades

Hydroxytyrosol grades run from a few percent in crude extracts to 99 percent or higher in isolated material.

Olive fruit and olive leaf extract grades typically contain 6 to 25 percent hydroxytyrosol, often blended with oleuropein, tyrosol, and verbascoside.

Concentrated grades at 40 to 80 percent are produced for capsule and softgel manufacture. Some suppliers offer water-soluble powders standardised to 20 or 40 percent on a maltodextrin or gum acacia carrier.

Synthetic hydroxytyrosol at 99 percent or higher is produced for pharmaceutical and reference use, with full impurity profiling. Liquid concentrates derived from olive mill wastewater are also available for beverage applications.

Regulatory Status

Hydroxytyrosol is approved as a novel food in the European Union under Commission Implementing Regulation (EU) 2017/2470, covering synthetic hydroxytyrosol for use in fats and supplements at defined levels.

The EFSA cardiovascular health claim authorised under Regulation (EU) No 432/2012 reads that olive oil polyphenols contribute to the protection of blood lipids from oxidative stress, conditional on the food providing at least 5 mg of hydroxytyrosol and its derivatives (such as oleuropein complex and tyrosol) per 20 g of olive oil. This is the only authorised polyphenol health claim in the EU referenced to a single compound family.

In the United States, olive fruit and olive leaf extracts standardised to hydroxytyrosol are marketed as dietary-supplement ingredients under DSHEA, with several self-affirmed GRAS notifications covering food use.

Pharmacopoeial monographs do not yet exist for the isolate, although olive leaf monographs in the European Pharmacopoeia and the United States Pharmacopeia define oleuropein-based identity and assay tests.

Manufacturing Process

Three main routes account for current hydroxytyrosol supply.

The dominant natural route is recovery from olive mill wastewater and olive pomace, the residual streams of olive oil pressing in Spain, Italy, Greece, and Tunisia. Hydroxytyrosol is concentrated in the aqueous phase and is extracted by adsorption on macroporous resin or by membrane separation, followed by acid or enzymatic hydrolysis of the parent oleuropein to release the free phenol.

A second route extracts olive leaf, which is rich in oleuropein. Hydrolysis under controlled acid or enzymatic conditions converts the secoiridoid into hydroxytyrosol and elenolic acid, which are then separated chromatographically.

The synthetic route condenses 3,4-dihydroxybenzaldehyde or 3,4-dimethoxyphenylacetic acid through standard reductive steps to yield racemic hydroxytyrosol. The synthetic material is identical to the natural compound, which lacks chirality.

Final material leaves the plant as a liquid concentrate, a powder on a food-grade carrier, or a crystalline solid depending on grade.
