L-Histidine Hydrochloride is the hydrochloric acid salt of the imidazole-bearing amino acid L-histidine, typically sold as the monohydrate crystal C₆H₉N₃O₂·HCl·H₂O.
The compound has a molar mass of 209.63 g/mol and CAS number 5934-29-2. It is the pharmacopoeial form of histidine, preferred over the free base because the salt has markedly higher water solubility and better storage stability.

USP and Ph. Eur. grades dominate sales volume. These are the grades used for parenteral nutrition amino acid blends, infant formula fortification, and biologic-drug formulation buffers, and they carry tighter limits on heavy metals and bacterial endotoxins.

Regulatory Status
L-Histidine Hydrochloride is recognised as a nutrient and pharmaceutical excipient in essentially every regulated market.
In the European Union Regulation (EC) 1925/2006 permits it for food fortification, and it also has approval as a food supplement ingredient. JECFA has not set a numerical ADI, since histidine is an essential nutrient for infants and a conditionally essential one for adults.
The U.S. Food and Drug Administration recognises L-Histidine as GRAS under 21 CFR 172.320 for use as a nutrient in foods. The hydrochloride salt form is the form normally traded.
Pharmacopoeial monographs appear in the United States Pharmacopeia (USP), the European Pharmacopoeia (Ph. Eur.), and the Japanese Pharmacopoeia (JP). The FCC monograph covers food-grade material.
Halal and Kosher certifications are routinely available, since fermentation feedstocks are plant-derived. Most producers also hold non-GMO documentation and allergen-free attestations on request.
Manufacturing Process
Commercial L-Histidine Hydrochloride is produced almost exclusively by fermentation.
The strain of choice is an engineered Corynebacterium glutamicum or Escherichia coli, carrying mutations that release feedback inhibition on the his operon. Broth titers of 30 to 60 g/L of free L-histidine are typical.
The fermentation feedstock is regional. Cane molasses dominates in Brazil and Southeast Asia, corn glucose syrup in North America and East Asia. After cells are removed by filtration, the broth is acidified to the isoelectric point and the free amino acid is recovered by ion-exchange chromatography.
The purified L-histidine is then dissolved in dilute hydrochloric acid, decolorised over activated carbon, and crystallised as the monohydrate hydrochloride salt. Crystals are washed, centrifuged, and dried under controlled humidity to preserve the water of crystallisation.
