Riboflavin Sodium Phosphate
Riboflavin Sodium Phosphate is the sodium salt of riboflavin 5'-monophosphate and the water-soluble form of Vitamin B2 used in clear beverages, parenteral nutrition, and any application where native riboflavin's poor solubility creates a formulation problem.
It is an orange to yellow-orange crystalline powder with the molecular formula C₁₇H₂₀N₄NaO₉P and a molar mass of 478.33 g/mol. Compared with the parent vitamin (water solubility around 80 mg/L), the sodium phosphate ester dissolves freely at roughly 100 g/L, which is the practical reason it exists as a separate commercial article.

The standard pharmaceutical grade meets USP, Ph. Eur., and JP monographs, with assay typically reported on an anhydrous and free-riboflavin-corrected basis. Total riboflavin phosphate esters run around 73 to 79 percent of the labelled value because the commercial product is a mixture of the 5'-monophosphate (dominant) with smaller amounts of other phosphate isomers and free riboflavin.
A food-grade fraction with the same chemical profile and a relaxed documentation package is produced for beverage fortification and pharmaceutical-equivalent supplement use.
Material is normally micronized to pass 100 mesh for fast dissolution in cold water without need for heating or mechanical shear.

In the European Union it carries the E number E101(ii), and Regulation (EC) 1333/2008 permits it. U.S. FDA approves it as a colour additive exempt from certification under 21 CFR 73.450 and as a nutrient under GRAS.
JECFA has set a group ADI for riboflavin and its phosphate esters together. Pharmacopoeial monographs appear in USP, Ph. Eur., and JP.
Halal and Kosher certifications are routinely available when the upstream riboflavin is produced on a plant-based fermentation medium.
Manufacturing Process
Riboflavin Sodium Phosphate is produced from riboflavin in a single phosphorylation step.
Fermentation-derived riboflavin, typically from a Bacillus subtilis or Ashbya gossypii process, is the starting material. The 5'-hydroxyl group is selectively phosphorylated using a phosphorus oxychloride or polyphosphoric acid reagent under controlled temperature and pH.
The crude product is a mixture of the desired 5'-monophosphate with smaller amounts of 4'- and 3'-isomers and unreacted riboflavin. The reaction mixture is quenched, neutralised with sodium hydroxide to form the sodium salt, and concentrated under vacuum.
Final purification is by precipitation from a controlled water-ethanol or water-acetone system. The product is filtered, washed, and dried to give the characteristic orange-yellow crystalline powder.
