Vitamin B1

Vitamin B1 is a water-soluble nutrient sold to the food, beverage, pharmaceutical, and feed industries in two main salt forms: thiamine hydrochloride (CAS 67-03-8) and thiamine mononitrate (CAS 532-43-4).

The molecule consists of a pyrimidine ring linked through a methylene bridge to a thiazolium ring. After phosphorylation in the body it becomes thiamine pyrophosphate, the cofactor required by pyruvate dehydrogenase, alpha-ketoglutarate dehydrogenase, and transketolase.

Common Markets Grades

Two salt forms cover almost all commercial demand.

Thiamine hydrochloride is the more soluble salt and is preferred for liquid pharmaceuticals, injectables, oral solutions, and effervescent tablets. It is hygroscopic and sold at 98.5 percent minimum assay against the USP and Ph. Eur. monographs.

Thiamine mononitrate has lower hygroscopicity and is the standard choice for dry fortification of flour, cereal, infant formula, and multivitamin tablets. It assays at 98 to 101 percent and offers better stability in dry premixes than the chloride salt.

Feed-grade thiamine mononitrate runs at 98 percent for poultry, swine, and aquaculture premixes. Pharmaceutical-grade material comes as fine powder or as a granulated direct-compression grade for tableting.

Regulatory Status

Thiamine is approved as a nutrient in essentially every regulated market.

In the European Union Regulation (EC) No 1925/2006 on the addition of vitamins and minerals to food lists it, as does Directive 2002/46/EC for food supplements. Both the hydrochloride and mononitrate forms are authorised.

The U.S. Food and Drug Administration recognises thiamine hydrochloride and thiamine mononitrate as GRAS under 21 CFR 184.1875 and 21 CFR 184.1878 respectively. Enrichment levels for flour and cereal products are set in 21 CFR 137.

Pharmacopoeial monographs appear in the USP, Ph. Eur., JP, and BP. EFSA has set tolerable intake guidance and confirmed no upper limit for oral intake based on the absence of identified adverse effects.

JECFA reviewed thiamine and did not assign a numerical limit, classifying it as a nutrient where intake is governed by reference dietary values rather than toxicological endpoints.

Industrial thiamine is made entirely by chemical synthesis. The total synthesis route developed by Williams and Cline in 1936 remains the basis for current manufacturing.

The pyrimidine ring is built from acetamidine and ethyl formylpropionate, then converted to the bromomethyl pyrimidine. In parallel, the thiazole half is prepared from carbon disulfide and chloroacetaldehyde routes that yield 4-methyl-5-hydroxyethylthiazole.

The two halves are coupled by N-alkylation of the thiazole nitrogen with the bromomethyl pyrimidine, generating the thiamine cation. Treatment with hydrochloric acid gives the hydrochloride salt; treatment with nitric acid gives the mononitrate.

Crude product is recrystallised from aqueous ethanol to pharmacopoeial purity. Global capacity is concentrated in a small number of Chinese producers, with smaller volumes from India and a residual European manufacturer.
