Manufacturing Process

Industrial Alpha-Ketoglutarate is produced primarily by microbial fermentation. Yeasts of the genus Yarrowia lipolytica and selected bacterial strains accumulate the keto-acid extracellularly when cultivated on glycerol, n-alkane, or rapeseed oil substrates under nitrogen-limited conditions.

After fermentation, biomass is removed by centrifugation or microfiltration, and the keto-acid is recovered through a combination of ion-exchange chromatography, electrodialysis, and crystallisation from aqueous solution.

A chemical synthesis route exists, proceeding from oxalacetate and acetate or from glutamic acid by oxidative deamination, but fermentation is now economically dominant at industrial scale.

Salt forms are produced by neutralising the free acid with the corresponding base, typically calcium hydroxide, sodium carbonate, or the relevant amino acid in aqueous solution, followed by crystallisation and drying.
