Vitamin B6

Vitamin B6 is a family of three interconvertible pyridine derivatives: pyridoxine, pyridoxal, and pyridoxamine. The commercial form is pyridoxine hydrochloride (CAS 58-56-0), sold to the food, supplement, pharmaceutical, and feed industries.

All three forms are converted in the body to pyridoxal 5-phosphate (PLP), the active cofactor for more than 140 enzymes including transaminases, decarboxylases, and the synthesis of heme, serotonin, and gamma-aminobutyric acid.

Common Markets Grades

Pyridoxine hydrochloride dominates commercial supply. It meets 98 to 101 percent assay against the USP, Ph. Eur., and JP monographs.

The standard fine-powder grade serves dietary supplements, multivitamin tablets, and pharmaceutical use. Granulated and direct-compression grades are produced for high-speed tableting and effervescent applications.

Feed-grade pyridoxine hydrochloride at 98 percent is produced for poultry, swine, dairy, and aquaculture premixes. Spray-dried and coated variants improve stability in mineral-rich premixes and extruded feeds.

Pyridoxal 5-phosphate (P5P) is a finished-form active used for premium supplement positioning. It runs at 98 percent assay as the monohydrate and commands a price premium over pyridoxine HCl based on the claim of bypassing hepatic phosphorylation, though the clinical relevance is modest in healthy adults.

Pyridoxamine dihydrochloride is produced at niche scale for research and specialty supplement use.

Regulatory Status

Vitamin B6 is approved as a nutrient in essentially every regulated market.

In the European Union Regulation (EC) No 1925/2006 authorises pyridoxine hydrochloride, pyridoxal 5-phosphate, and pyridoxamine dihydrochloride for food fortification, and Directive 2002/46/EC authorises them for food supplements. EFSA reviewed the upper intake level in 2023 and reduced it to 12 mg/day for adults, reflecting evidence on peripheral neuropathy at chronic high intake.

The U.S. Food and Drug Administration recognises pyridoxine hydrochloride as GRAS under 21 CFR 184.1676. The Institute of Medicine has set a tolerable upper intake level of 100 mg/day in adults.

JECFA has reviewed pyridoxine and assigned it acceptable nutrient status. Pharmacopoeial monographs appear in the USP, Ph. Eur., JP, and BP.

Halal and Kosher certifications are widely available given the fully synthetic supply chain. Cosmetic use of pyridoxine and its derivatives is listed in the EU CosIng database.

Manufacturing Process

Industrial pyridoxine hydrochloride is produced by chemical synthesis. Several routes have been operated commercially, all building the pyridine ring with the correct substitution pattern of hydroxyl, hydroxymethyl, and methyl groups.

The most widely used route is the Diels-Alder approach pioneered by Merck and refined by Daiichi and several Chinese producers. 4-methyl-5-alkoxyoxazole is reacted with a dienophile such as dimethyl maleate or 2-butene-1,4-diol, and the bicyclic intermediate is opened and aromatized to the pyridoxine skeleton.

The product is acidified with hydrochloric acid to give the crystalline hydrochloride salt, recrystallised from aqueous ethanol, and dried to pharmacopoeial purity. For pyridoxal 5-phosphate, pyridoxine is oxidised to pyridoxal and then phosphorylated chemically or enzymatically.

Production is concentrated in a small number of Chinese plants, with smaller volumes from India. Pyridoxine is one of the more synthesis-intensive B vitamins, and the cost structure reflects multiple stages and stringent quality control.
