Manufacturing Process

Industrial 2-Methyl-3-Tetrahydrofuranthiol is produced by catalytic hydrogenation of 2-methyl-3-furanthiol or by independent synthesis from saturated building blocks.

Selective hydrogenation of the furan ring of 2-methyl-3-furanthiol under controlled hydrogen pressure with a palladium or nickel catalyst gives the tetrahydrofuran product directly. Reaction conditions must be tuned to spare the thiol from desulfurization, which is the main side reaction.

An alternative route starts from 2-methyltetrahydrofuran-3-one. Reduction of the ketone to the corresponding alcohol followed by conversion to the thiol (for example through a thioacetate intermediate and hydrolysis) delivers the target molecule in defined stereochemistry.

Crude product is purified by vacuum distillation under nitrogen and stabilised with a trace antioxidant. The finished liquid is packed under inert gas to limit oxidative dimerization on storage.
