Broccoli Extract is a vegetable-derived concentrate prepared from the florets and aerial parts of Brassica oleracea var. italica. It is standardised on glucosinolates (primarily glucoraphanin) and, in active grades, on the bioactive isothiocyanate sulforaphane.

Common Markets Grades
Broccoli extract from mature plant material is graded by either glucoraphanin content or by active sulforaphane content.
Glucoraphanin-standardised florets-and-stem grades sit at 0.3, 1, 2, and 5 percent glucoraphanin. The 1 percent grade is the standard grade in low-cost broccoli capsule lines.
Sulforaphane-active grades, in which myrosinase is preserved or co-formulated to deliver the bioactive isothiocyanate on contact with water, are specified by milligrams of sulforaphane yield per gram. Mainstream grades deliver 0.1 to 0.5 percent active sulforaphane.
Polyphenol and antioxidant grades at 5 to 20 percent total phenolics are made for general antioxidant formulas.
Whole-vegetable broccoli powders without standardised actives are sold to the greens-powder and superfood category.
Material is typically a fine green to brownish-green spray-dried powder with a characteristic cruciferous note. Sulforaphane-active grades often carry a moisture-barrier coating to prevent premature myrosinase activation.

Regulatory Status
Broccoli is a globally consumed vegetable and its extracts are widely recognised as food-derived ingredients.
In the United States broccoli extract and broccoli sprout extract are sold as dietary-supplement ingredients under DSHEA. Multiple commercial preparations carry self-affirmed GRAS opinions for use in functional foods and beverages.
In the European Union dried broccoli and conventional broccoli extracts have a pre-1997 history of food use. Concentrated sulforaphane or glucoraphanin preparations produced by novel processes may require novel-food assessment.
In Japan broccoli sprout preparations have a long history of use and several products have cleared the Foods with Function Claims notification system.
No specific JECFA evaluation applies to broccoli extract, which is treated as a food.
Manufacturing Process
Broccoli for extraction is sourced from contracted growers in China, the United States, Mexico, and Europe. Florets, stems, and aerial parts are used depending on the target specification.
For glucoraphanin-standardised grades the harvested material is rapidly blanched to inactivate native myrosinase, then dried, milled, and extracted with hot water or aqueous ethanol.
The crude extract is filtered, concentrated under vacuum, and either spray-dried directly or passed through anion-exchange resin to enrich the glucosinolate fraction.
For sulforaphane-active grades the manufacturing strategy is different. Myrosinase activity is preserved by gentle drying below 50 degrees C, or a separate myrosinase concentrate is co-formulated with a glucoraphanin-rich powder. Activation occurs when the consumer adds water at use.
Whole-vegetable powders are produced by simple drying and milling, without standardisation.
Glucoraphanin and other glucosinolates are quantified by HPLC after desulfation. Sulforaphane is measured by HPLC after enzymatic conversion, with cyclocondensation assays as an alternative.
