Casein Phosphopeptides, abbreviated CPP, are a family of phosphorylated peptides released from milk casein by enzymatic digestion. They are best known for their ability to bind and stabilise calcium and phosphate ions in amorphous, bioavailable form.
The defining structural feature is a cluster of phosphoserine residues separated by glutamic-acid residues, often summarised as the SerP-SerP-SerP-Glu-Glu motif. This cluster acts as a multidentate ligand for calcium and is the basis for nearly every application of CPP in food and oral care.

Standard food and supplement grades are off-white to pale yellow powders at a minimum CPP content of 80 percent, with the balance composed of non-phosphorylated peptides and minor lactose and ash.
Oral-care grades are CPP-ACP, a complex of casein phosphopeptide with amorphous calcium phosphate. This complex is the active ingredient in remineralizing toothpastes, dental crèmes, and chewing gums.
A CPP-ACFP variant, in which fluoride is co-stabilised with the calcium phosphate cluster, is produced for fluoride-containing dental products.

Casein phosphopeptides are accepted as food and supplement ingredients in major markets when produced from food-grade casein under sanitary conditions.
In the United States the ingredient operates under GRAS practice for use in foods, beverages, and dietary supplements. The CPP-ACP complex used in oral-care products is regulated through dental-product channels rather than food channels.
In the European Union, CPP is treated as a dairy-derived ingredient and is subject to milk-allergen labelling. Use in food contact and supplement applications follows the standard rules for hydrolysed milk-protein ingredients.
Halal and Kosher certifications are routinely available because the starting casein and the processing aids are compatible with both standards when properly sourced.
Manufacturing Process
Manufacture begins with bovine casein, either as acid casein or as caseinate. The casein is dissolved and subjected to controlled enzymatic hydrolysis with trypsin or a similar pancreatic protease.
The hydrolysate is then enriched in phosphopeptides by selective precipitation. Addition of calcium and ethanol or pH adjustment causes the phosphorylated peptides, which bind calcium tightly, to drop out of solution while the non-phosphorylated background remains soluble.
The phosphopeptide pellet is washed, redissolved, and may be passed over an ion-exchange or membrane step for further enrichment. Ultrafiltration removes residual large peptides and proteins.
The purified concentrate is dried, usually by spray drying, and blended to a target phosphopeptide content. CPP-ACP variants are produced by combining the dried CPP with calcium and phosphate ions under controlled pH to form the amorphous complex before final drying.
