Copper Bisglycinate is a chelate of copper with two molecules of the amino acid glycine. It has the formula Cu(C2H4NO2)2 and CAS number 13479-54-4. The compound contains approximately 24 percent elemental copper.

The chelate is a fine blue powder of moderate water solubility. Its bonded structure shields the copper ion from binding with dietary phytate, polyphenols, and competing minerals in the gut, supporting the highest bioavailability tier among commercial copper forms.

Copper Bisglycinate serves nutrition channels as a chelated copper source.

Fully reacted bisglycinate grade carries two glycine ligands per Cu(II) centre. Chelation is verified by infrared spectroscopy, which shows the disappearance of the free glycine carboxyl stretch and the appearance of the metal-bonded carboxylate band.

Pharmaceutical and supplement grade is graded at 18 to 24 percent elemental copper depending on water content. Heavy metal limits are aligned with ICH Q3D Option 2A for oral supplements.

Infant nutrition grade is produced with tight microbiological limits, low residual solvents, and full traceability of glycine origin.

Spray-dried agglomerated grade is produced for direct-compression tablets and capsule blending. Fine powder grade is preferred for liquid suspensions and powder beverage mixes.

Regulatory Status

Copper Bisglycinate is approved as a nutrient source across major global markets.

The European Union authorises Copper Bisglycinate as a source of copper for food supplements under Annex II of Directive 2002/46/EC and for fortified foods under Regulation (EC) No 1925/2006. The compound was added to the permitted list following a positive EFSA scientific opinion on its safety and bioavailability.

The U.S. Food and Drug Administration recognises amino acid chelates of copper, including copper bisglycinate, as suitable for use in dietary supplements under the general framework for nutrient sources.

EFSA scientific opinion (EFSA Journal 2009;7(11):1383) concluded that copper bisglycinate is bioavailable and safe at intended use levels in food supplements.

For use in infant formula, copper compounds including the bisglycinate are evaluated case by case against Regulation (EU) 2016/127. Halal, Kosher, vegan, and non-GMO certifications exist when the glycine source qualifies.

Manufacturing Process

Copper Bisglycinate is manufactured by direct chelation of copper ions with glycine in aqueous solution.

Glycine is dissolved in deionized water at 50 to 70 degrees Celsius to form a 10 to 20 percent solution. Copper carbonate (basic) or freshly precipitated copper hydroxide is added in two-to-one molar ratio of glycine to copper.

The reaction is held with gentle agitation for 1 to 3 hours under mild heat. The pH stabilises in the 6.5 to 7.5 region as the glycine carboxyl groups deprotonate and the amine nitrogen and carboxyl oxygen form coordinate bonds with the copper centre. The mixture turns from pale slurry to a deep blue solution as the chelate forms.

Infrared spectroscopy is used inline or on samples to confirm chelation. The clear blue solution is filtered to remove any unreacted copper source, concentrated under vacuum, and spray-dried or allowed to crystallise on cooling.

The dried product is milled and tested by ICP-MS for heavy metals and by infrared spectroscopy for chelation degree. Each batch is also tested for free glycine, residual copper carbonate, and microbiological compliance.
