Zinc Bisglycinate
Zinc bisglycinate is a zinc amino acid chelate in which a zinc ion is held by two glycine molecules, each binding through its amino nitrogen and a carboxyl oxygen to form two stable five membered rings around the metal.
The material is a white to off white free flowing powder. Fully reacted anhydrous chelate has a theoretical zinc content near thirty percent, and commercial grades are declared between about twenty and thirty percent depending on hydration and on whether free glycine has been left in the product as a buffer. Formulators dose on the declared elemental zinc figure rather than on the chelate assay.
Solubility in water is modest at neutral pH and increases in acid conditions, so the powder disperses in dry blends and premixes without dissolving and passes into solution in the stomach. The chelate is less astringent and less metallic in taste than zinc sulphate, which matters in chewables and powders, and it interacts less aggressively with vitamins in a premix, giving better vitamin C and vitamin A retention over shelf life.

Food and supplement grade is the main tier, made to a controlled two to one glycine to zinc ratio, declared on elemental zinc, and tested for heavy metals and microbiology for use in tablets, capsules, powders and fortified foods. Feed grade chelate is the higher volume product, specified on guaranteed zinc content for trace mineral premixes and complete feeds.
Within the food tier, the practical choices are assay and physical form. High assay grades close to the stoichiometric zinc limit are used where dose volume is limited, and buffered grades carrying additional free glycine are preferred where a lower, easier to blend assay suits the premix. Regulatory Status
In the United States zinc bisglycinate is used as a dietary ingredient in supplements manufactured under 21 CFR Part 111, and the chelate has also been the subject of a GRAS notification supporting defined uses in conventional foods and beverages, alongside the long established zinc sources such as zinc sulphate, oxide and gluconate that 21 CFR Part 182 lists as generally recognised as safe. For animal feed, zinc amino acid chelates are covered by the Association of American Feed Control Officials ingredient definitions.
In the European Union the annex to Directive 2002/46/EC sets out the zinc forms permitted in food supplements, and Regulation 1925/2006 sets out those permitted for the fortification of foods, both of which have been amended over time to add amino acid chelate forms following EFSA opinions, so formulators confirm the current listing for the exact chelate before use. Zinc chelate of glycine hydrate is separately authorised as a nutritional feed additive in the EU feed additive register with its own purity conditions. Codex addresses zinc through the advisory lists of nutrient compounds attached to its infant formula and special dietary use standards.
Manufacturing Process
The chelate is formed by reacting a zinc source with glycine in water. Zinc oxide or zinc carbonate is slurried with two molar equivalents of glycine and heated with agitation; the reaction is driven by consumption of the basic zinc compound and, in the carbonate case, by the escape of carbon dioxide. Using the oxide or carbonate rather than the sulphate avoids leaving a mineral anion in the product, which would otherwise have to be washed out and would show up in the sulphated ash result.
The mixture is held at controlled temperature and pH until chelation is complete, verified by the fall in free glycine and by infrared confirmation of the chelate bond. Unreacted zinc solids are removed by filtration, and the liquor is concentrated and dried, most often by spray drying, which gives a rounded, free flowing particle. Where a denser particle is wanted the wet cake is dried in a fluid bed instead. The dried solid is milled, sieved, passed over magnets and through a metal detector, then blended to the declared zinc content and packed.
