Hesperitin is a citrus flavanone aglycone (CAS 520-33-2) recognised for its role as the active metabolite of the more widely consumed glycoside hesperidin.
The compound has the formula C₁₆H₁₄O₆ and a molar mass of 302.28 g/mol. It is a pale yellow crystalline powder, sparingly soluble in water and freely soluble in dimethyl sulfoxide, ethanol, and dilute alkali.

Standard food-supplement grade ranges from 90 to 95 percent assay by HPLC, recovered from enzymatic or acidic hydrolysis of hesperidin sourced from sweet orange (Citrus sinensis) peel.
A 98 percent pharmaceutical grade is produced for solid-dose formulation and clinical research, with tight controls on residual hesperidin, isosihesperidin, and solvent residues.
Reference material at 99 percent or higher is produced for analytical and pharmacopoeial calibration. Micronized and amorphous solid dispersion variants exist where dissolution rate is the limiting factor.

Regulatory Status
Hesperitin is not a standalone food additive. It is present as a minor constituent of permitted citrus extracts and citrus juice, with intake covered by general food law.
In the European Union it falls under Regulation (EC) No 178/2002 as a food constituent and is sold to manufacturers as a botanical extract or purified ingredient. No novel-food authorisation is required when sourced from edible citrus species at normal intake levels.
In the United States, hesperitin-containing citrus extracts are marketed as dietary-supplement ingredients under DSHEA. The aglycone itself has been the subject of self-affirmed GRAS notifications by individual suppliers.
EFSA has evaluated a related health claim on hesperidin and orange juice flavanones for endothelial function and issued an unfavorable opinion in 2010, citing insufficient evidence. No authorised EU health claim currently applies to hesperitin.
Pharmacopoeial monographs do not yet exist, although Chinese Pharmacopoeia entries cover related citrus flavonoids used as quality markers.
Manufacturing Process
Industrial hesperitin is obtained almost exclusively by hydrolysis of hesperidin.
Hesperidin itself is recovered from the peel, pith, and segment membranes of Citrus sinensis, Citrus reticulata, and Citrus aurantium, which are byproducts of orange juice processing in Brazil, Mexico, Spain, and Italy.
The glycoside is cleaved by either dilute mineral acid at elevated temperature or by α-L-rhamnosidase and β-D-glucosidase enzymes. Enzymatic hydrolysis offers cleaner isomer profiles and lower residual sugars but at higher unit cost.
The crude aglycone is then recrystallised from aqueous ethanol or methanol. Polishing chromatography on macroporous resin upgrades the material to pharmaceutical and reference-standard purities.
A small share of supply comes from semi-synthesis starting from naringenin via 3'-hydroxylation and 4'-O-methylation, used mainly where isotope-labelled or structurally modified hesperitin is required for research.
