Rutin

Rutin is a flavonol glycoside, chemically quercetin-3-O-rutinoside, with formula C27H30O16 and molar mass 610.52 g/mol for the anhydrous compound and 664.56 g/mol for the trihydrate that most commercial material is. It consists of the aglycone quercetin bearing the disaccharide rutinose, made up of rhamnose and glucose, at the 3 position.

It occurs widely in plants and is concentrated in the flower buds of Sophora japonica, in buckwheat herb and in citrus peel, the first of these being the dominant industrial source. Commercially it serves two purposes: as an antioxidant dietary ingredient in its own right, positioned on capillary and circulatory support, and as the feedstock from which quercetin, isoquercitrin and enzymatically modified isoquercitrin are made.

The material is a pale yellow to greenish yellow crystalline powder, only slightly soluble in cold water and better in hot water, soluble in ethanol and methanol and freely soluble in dilute alkali. That limited aqueous solubility is the reason the more soluble derivatives exist. Rutin is stable in the dry state and to normal food processing but darkens and oxidises in alkaline solution and is degraded by prolonged light exposure, so it is packed away from light and moisture.

Common Markets Grades

The standard article is rutin at 95 percent minimum by ultraviolet assay or by HPLC, sold as the trihydrate, with 98 percent grades traded for pharmaceutical and reference use. The specification states whether assay is on the dried or the as-is basis, since the hydrate water is a significant fraction of the weight. Lower assay grades at 80 and 90 percent are sold for feed and for further processing.

Derivative grades are commercially as important as rutin itself. Troxerutin, the hydroxyethylated derivative, and isoquercitrin, produced by enzymatically removing the rhamnose, are both traded as separate articles, and enzymatically modified isoquercitrin, in which glucose units are transferred onto isoquercitrin to give a far more water soluble product, has its own specification and regulatory position. Physical forms are crystalline powder at 80 mesh, micronised material where dissolution rate matters, and water dispersible versions on a modified starch carrier. Source is stated on the certificate, since Sophora derived and buckwheat derived material differ in their impurity profile.

Regulatory Status

In the United States rutin is marketed as a dietary ingredient under the Dietary Supplement Health and Education Act and has a long history of use in bioflavonoid supplements. There is no food additive regulation and no colour additive listing for rutin itself, so its use in conventional foods rests on a GRAS conclusion for the specific preparation. Enzymatically modified isoquercitrin, made from rutin, has been the subject of a GRAS notice to the Food and Drug Administration for use as an antioxidant, to which the agency responded without objection, and that derivative therefore has a clearer food position than rutin does.

In the European Union rutin carries no E number and is not an authorised food additive. Its use in food supplements is handled at member state level under national rules on substances other than vitamins and minerals. Enzymatically modified isoquercitrin has been authorised as a novel food under Regulation 2015/2283 and has also been evaluated as an antioxidant, again giving the derivative a more defined status than the parent glycoside. The Joint FAO WHO Expert Committee has evaluated enzymatically modified isoquercitrin among flavonoid preparations. Regulation 1223/2009 governs cosmetic use, and Codex has no commodity standard covering rutin.

Manufacturing Process

Dried flower buds of Sophora japonica, or dried buckwheat herb, are cleaned and milled to a coarse powder. The material is extracted with hot water or with dilute alkaline water at 80 to 95 degrees Celsius over several counter current stages, since rutin dissolves far better hot than cold and better still with a small addition of lime or sodium hydroxide. The liquor is separated from spent solids on a decanter centrifuge and screened.

The hot liquor is clarified by plate filtration and then cooled slowly and adjusted to slightly acid pH, at which point crude rutin crystallises out because its solubility falls sharply on cooling and on neutralisation. The crude crop is recovered on a centrifuge and washed. It is then redissolved in hot aqueous ethanol or in dilute alkali, treated with activated carbon to remove colour and oxidation products, filtered hot, and recrystallised by controlled cooling, a cycle that is repeated where a 98 percent grade is required. The wet cake is centrifuged, washed with cold alcohol and water, dried under vacuum at low temperature to preserve the hydrate, milled to the specified mesh, assayed and packed under a moisture and light barrier.
