Mucuma Extract

Mucuma extract, more correctly written mucuna extract, is a concentrated preparation of the seed of Mucuna pruriens (L.) DC., the velvet bean, a tropical legume grown across India, Southeast Asia, Africa and Central America. The spelling with an m is a transliteration variant that appears widely in the ingredient trade and refers to the same species.

The extract is standardised on levodopa, also written L-DOPA, which is 3,4-dihydroxy-L-phenylalanine, a non-protein amino acid and the direct metabolic precursor of dopamine. Velvet bean seed accumulates levodopa at unusually high levels for a plant, commonly three to seven percent of the dry seed, and this is the sole basis on which the extract is specified and priced. The seed also carries tryptamine alkaloids, serotonin, nicotine and beta-carboline compounds at trace levels, along with the ordinary legume constituents of starch, protein and fibre.

The extract is a light brown to grey-brown fine powder with a faint beany odour and a slightly bitter taste. Levodopa is only sparingly soluble in water and dissolves better in dilute acid; it is markedly unstable in solution at neutral and alkaline pH, where it oxidises rapidly to dopaquinone and then to dark melanin-like polymers, which is why extracts darken visibly if they take up moisture or are held in alkaline conditions. Commercial powder is packed with a moisture barrier, kept away from light and often stabilised with ascorbic acid or citric acid in formulation.

Common Markets Grades

Grades are quoted on levodopa content by HPLC. Fifteen and twenty percent are the common commodity levels, 40, 60 and 98 percent grades exist and approach the isolated compound at the top end. Ratio extracts at 4:1 or 10:1 without a marker claim and milled seed powder, which carries the native three to seven percent, are traded alongside the standardised grades and are much cheaper. Buyers should note that some suppliers quote figures obtained by ultraviolet spectrophotometry, which returns higher numbers than HPLC on the same material, so the analytical method is part of the specification.

Extraction solvent is stated, since water and dilute acid systems and alcoholic systems differ in what they co-extract. Whether the seed coat has been removed matters, because the itching hairs on the pod and the seed coat constituents are the source of the plant's common name and are excluded from food-grade material. Origin is specified, with Indian material dominating supply. High assay grades above 60 percent are close enough to a purified compound that residual solvent and single impurity limits become the specifications that matter rather than extract ratio.

Regulatory Status

Mucuna pruriens occupies an awkward regulatory position because its active constituent is also a prescription pharmaceutical. In the United States the seed and its extracts are marketed as dietary ingredients under the Dietary Supplement Health and Education Act, with manufacture under 21 CFR Part 111, but levodopa itself is an approved drug substance, and the Food and Drug Administration has taken the position that a substance approved as a drug cannot be marketed as a dietary supplement ingredient. In practice the agency has treated botanical extracts standardised on naturally occurring levodopa differently from the isolated synthetic compound, but a high assay extract approaching the purity of the drug substance carries real regulatory risk in that market.

In the European Union Mucuna pruriens seed has no significant history of food consumption before May 1997 and falls within the scope of the novel food Regulation, so extracts require authorisation before being placed on the market as food or food supplement ingredients. Several member states additionally restrict levodopa-containing preparations on the grounds that the substance is pharmacologically active and is a medicinal ingredient. No health claim for mucuna appears on the EU register of authorised claims. Neither Codex nor JECFA has evaluated the material or published a specification, so trade relies on supplier monographs and on the analytical data reported with each lot.

Manufacturing Process

Production begins with dried velvet bean seed, cleaned to remove pods, stones and foreign seed, then dehulled where the grade requires it and milled to a coarse meal. Levodopa is extracted with water acidified to around pH 2 to 3, or with dilute aqueous ethanol containing an acid, because the amino acid is far more soluble and far more stable in acid than at neutral pH. Extraction is run at moderate temperature and completed promptly, since prolonged heating oxidises the target.

The liquor is filtered to remove starch and seed fibre, then clarified. Enrichment is by ion exchange: because levodopa is amphoteric, the acidified liquor is passed over a cation exchange resin that binds the amino acid while sugars, starch and neutral constituents pass through, and it is then eluted with dilute ammonia or a buffered salt solution. That step accounts for most of the difference between a 20 percent and a 60 percent grade. The eluate is adjusted back to acid pH and concentrated under vacuum at low temperature, with an antioxidant such as ascorbic acid or sodium metabisulphite added to suppress oxidation during processing. For the highest grades the concentrate is crystallised at the isoelectric point, filtered and recrystallised. The concentrate or crystals are dried under vacuum or spray dried at a low inlet temperature, standardised by blending or with a carrier, sieved and packed with a moisture and light barrier.
