Nucleotide
Nucleotide, as supplied for infant formula and adult nutrition, is a defined blend of the five 5'-monophosphates: adenosine (AMP), cytidine (CMP), guanosine (GMP), uridine (UMP), and inosine (IMP).
The blend approximates the nucleotide profile of human milk and is used to support the dietary pool available for rapidly dividing tissues, including the developing immune system and gut mucosa.

Individual sodium salts of AMP, CMP, GMP, UMP, and IMP run at 98 percent or higher purity. They are sold to formulators who blend in-house to their own ratio.
The standard infant-formula blend is a five-component mix with a ratio close to that of human milk, typically delivering total nucleotide at a few milligrams per 100 kcal in the finished formula.
A simpler four-component blend without IMP is produced for markets that follow that nutritional reference.
Adult supplement blends often include higher relative IMP for sports recovery positioning, where inosine pathway support is the reason for the formulation.

5'-Nucleotides are recognised in the Codex Alimentarius standard for infant formula as permitted optional ingredients, with the five-component blend reflecting the human-milk profile.
In the European Union, nucleotides are permitted for addition to infant formula and follow-on formula under the relevant Commission Delegated Regulation, with maximum levels set for each individual nucleotide.
In the United States, the individual 5'-nucleotides have GRAS status for use in infant formula and general food applications.
JECFA has reviewed the safety of nucleotide addition to infant formula at the levels relevant to commercial use.
Pharmacopoeial specifications for the individual sodium salts appear in the Japanese Pharmacopoeia and as reference monographs in food-additive compendia. Halal and Kosher certifications are routinely available.
Manufacturing Process
Industrial production of the individual 5'-nucleotides is dominated by microbial fermentation, with separate processes for the purine and pyrimidine series.
For the purine route, engineered Corynebacterium ammoniagenes or Bacillus subtilis strains are grown on glucose with controlled feeding to overproduce inosine or guanosine. The nucleoside is then phosphorylated chemically or enzymatically to yield the 5'-monophosphate.
For the pyrimidine route, fermentation strains overproducing orotic acid or uridine serve as the starting point. Conversion to UMP and CMP uses enzymatic phosphorylation followed by purification.
Downstream processing for all five nucleotides uses cell removal, activated carbon decolorisation, ion exchange, and crystallisation. The crystalline sodium salts are dried, milled, and sieved to a defined particle size.
Blending to the target five-nucleotide ratio is done in a dedicated clean area to maintain the homogeneity required for low-inclusion infant-formula use.
