Phloretin
Phloretin (CAS 60-82-2) is a dihydrochalcone polyphenol concentrated in apple peel, apple seeds, and the bark of various rosaceous trees.
The compound has the formula C₁₅H₁₄O₅ and a molar mass of 274.27 g/mol. It is a pale yellow crystalline powder, sparingly soluble in water and readily soluble in ethanol, propylene glycol, and most polar organic solvents.

Cosmetic-grade phloretin is typically sold at 90 to 95 percent assay, recovered from apple (Malus domestica) processing byproducts and from Phloridzin-rich Rosaceae bark.
A 98 percent grade is used in serums and dietary supplements where the highest stated purity is wanted. Reference and pharmaceutical grades at 99 percent or higher are produced with full impurity profiling, including limits on phloridzin, naringenin chalcone, and residual solvent.
Glycosylated and acetylated derivatives such as phloretin glucoside and phloretin acetate are sometimes offered alongside the aglycone for formulation flexibility.

Regulatory Status
Phloretin is a natural constituent of apple fruit and apple-derived foods, and intake from foods is governed by general food law.
In the European Union, phloretin is treated as a botanical-derived ingredient under Regulation (EC) No 178/2002 when sold as part of standardised apple extracts. It also appears on the European cosmetic ingredient inventory (CosIng) for use in finished cosmetic products under Regulation (EC) No 1223/2009.
In the United States, phloretin is widely used in cosmetic skincare products marketed under FDA cosmetic regulations and is included in several self-affirmed GRAS dossiers for apple polyphenol extracts.
No authorised EU health claim references phloretin specifically. Pharmacopoeial monographs for the pure compound do not yet exist, but pharmacopoeial apple-leaf and apple-peel reference texts are under development in several jurisdictions.
For cosmetic use, the SCCS has not flagged safety concerns at typical leave-on use levels of 0.1 to 2 percent.
Manufacturing Process
Industrial phloretin comes mainly from hydrolysis of phloridzin.
Phloridzin (phloretin-2'-O-glucoside) is recovered from apple processing residues, apple seedling root bark, and the bark of selected pear and cherry rootstocks. China and central Europe supply most of the raw material.
The glycoside is cleaved by β-glucosidase enzyme or by dilute acid at elevated temperature to release the free dihydrochalcone. Enzymatic hydrolysis is preferred for cosmetic grades because it minimises side products and residues.
The crude phloretin is recrystallised from aqueous ethanol or methanol. Activated carbon polishing, ion-exchange treatment, and final preparative chromatography deliver pharmaceutical and reference-grade material.
A small synthetic share is produced by hydrogenation of naringenin chalcone or by Friedel-Crafts acylation of phloroglucinol with 4-hydroxyphenylpropionic acid derivatives. Synthetic material is identical to the natural compound, which lacks chirality.
