Manufacturing Process

Commercial S-Adenosyl-L-Methionine is produced exclusively by yeast fermentation, since chemical synthesis cannot deliver the required optical purity at usable cost.

The strain of choice is Saccharomyces cerevisiae, typically a high-producing brewing or sake-yeast derivative. Fermentation is carried out aerobically on a glucose or molasses feedstock with L-methionine supplementation at the late log phase, which dramatically boosts intracellular SAMe accumulation.

At harvest, the yeast cells contain 1 to 4 percent SAMe on dry weight. Cells are lysed by acid extraction at low temperature, the SAMe is captured on a cation-exchange resin, eluted, and converted to the disulfate tosylate salt by sequential addition of sulphuric acid and p-toluenesulfonic acid.

The salt is crystallised from a water-ethanol system at low temperature, washed with cold ethanol, vacuum-dried below 30 °C, and packed under nitrogen with desiccant. The entire downstream process is kept at low temperature and short residence time to limit decomposition to S-adenosylhomocysteine.
