Allicin

Allicin is the principal sulphur-containing defense compound of crushed garlic and the molecule responsible for the bulb's sharp pungency and most of its reputed biological activity.

It is a thiosulfinate with the formula C₆H₁₀OS₂ (diallyl thiosulfinate). The compound does not exist in intact cloves. It is generated within seconds when the cell-wall enzyme alliinase contacts its substrate alliin after mechanical damage.

Allicin is highly unstable in pure form, so the commercial trade deals almost entirely in stabilised or assayed preparations rather than the free molecule.

The most common supplement-grade material is garlic powder standardised to a guaranteed allicin yield, typically 0.6 to 1.3 percent by enzymatic potential when reacted with water under defined conditions.

Enteric-coated tablet grades are formulated to bypass gastric acid, since allicin precursors degrade rapidly below pH 3.5. These are graded on an allicin-release basis verified by dissolution testing.

Liquid garlic extracts stabilised in glycerin or propylene glycol carry allicin contents in the 1000 to 5000 ppm range and are used by topical and veterinary formulators.

Regulatory Status

Allicin itself is not separately listed by most food authorities because it is treated as a constituent of garlic rather than an isolated additive.

Garlic and garlic preparations are GRAS in the United States under 21 CFR 182.10 and 182.20 when used as a spice, natural seasoning, or flavouring.

The European Food Safety Authority has reviewed garlic preparations and standardised garlic powder under the novel food and food supplement frameworks. Several allicin-yield-standardised extracts hold national authorisations in Germany under the Commission E monograph for Allium sativum.

Pharmacopoeial monographs for garlic powder appear in the European Pharmacopoeia, the United States Pharmacopeia, and the Japanese Pharmacopoeia, each with assay limits expressed as allicin or alliin equivalents.

For veterinary feed use, garlic and allicin-standardised preparations are listed as sensory feed additives in the EU register and as flavouring agents in the U.S. AAFCO framework.

Manufacturing Process

Industrial allicin is almost never isolated as the pure compound. Production focuses on generating and preserving it in matrix form.

The standard route begins with fresh garlic (Allium sativum) bulbs that are peeled, sliced, and freeze-dried or hot-air dried under temperature control to keep alliin and alliinase active in separate cellular compartments. The dried material is milled to a free-flowing powder and tested for enzymatic allicin yield.

For tablet manufacture, the powder is compressed and coated with an enteric polymer such as hydroxypropyl methylcellulose phthalate. The coating ensures alliinase and alliin meet only after gastric transit, releasing allicin in the small intestine.

Liquid extracts are prepared by macerating fresh garlic in cold water, allowing the enzymatic reaction to complete, then stabilising the filtrate in a glycerin or oil carrier at low temperature.

Synthetic allicin for research use is prepared by oxidation of diallyl disulfide with hydrogen peroxide or peracids in cold methanol, followed by silica chromatography under inert atmosphere.
