Bromelain

Bromelain is a mixture of sulfhydryl proteases recovered from the stem and fruit of the pineapple plant, Ananas comosus.

The preparation hydrolyses a broad range of peptide bonds and is one of the most widely used plant proteases in meat processing, brewing, and nutraceutical formulation.

Common Markets Grades

Bromelain is graded by activity rather than by mass.

Food-grade stem bromelain is graded at 1200 to 2400 GDU/g (gelatin digesting units) for meat tenderizing, beer chill-haze removal, and protein hydrolysate manufacture.

High-activity nutraceutical grades at 2400 GDU/g and above are spray-dried with maltodextrin or starch carriers for capsules, tablets, and joint-health stack formulations.

Fruit bromelain at 800 to 1500 BTU/mg is a separate gene product with a different substrate preference. It is produced for cosmetic peeling formulations and topical wound care.

Pharmaceutical grades are concentrated, lyophilized, and standardised to FIP units, with low endotoxin specification for oral and topical drug use.

Regulatory Status

JECFA recognises bromelain as a food enzyme with an ADI not specified.

The EU lists bromelain as a permitted food enzyme. The U.S. FDA accepts stem bromelain as GRAS under multiple notices for meat tenderizing, brewing, and protein hydrolysis applications.

Bromelain holds pharmacopoeial monographs in the United States Pharmacopeia and the European Pharmacopoeia for pharmaceutical and supplement use.

Halal and Kosher certifications are routinely available because the source is botanical. Pineapple allergen statements are required on retail-pack supplements and topical formulations.

Manufacturing Process

Industrial bromelain is recovered from pineapple processing waste. Pressed stems and rinds from juice plants in Thailand, the Philippines, Indonesia, and Costa Rica supply the global feedstock.

The plant material is crushed and extracted in cold buffered water. The clarified juice is concentrated by ultrafiltration to retain the 23 to 33 kDa enzyme fraction while removing sugars, polyphenols, and small peptides.

Cold acetone or ammonium sulphate precipitation pulls the protein out of solution. The pellet is redissolved, dialyzed, and lyophilized into the dry powder. Activity is checked against gelatin or casein standards before packing.

Cysteine and ethylenediaminetetraacetic acid are commonly added at low levels to keep the catalytic thiol reduced and to scavenge trace metals that would otherwise inactivate the enzyme during storage.
