Copper Chlorophyll

Copper Chlorophyll is the fat-soluble derivative of natural chlorophyll in which the central magnesium ion of the porphyrin ring has been substituted with a copper ion to give a far more stable and intensely green pigment.

The material carries the European food colour number E141(i) and is one of the two regulated copper-chlorophyll forms, and the second is the water-soluble sodium copper chlorophyllin coded E141(ii).

The copper substitution shifts the absorption maxima slightly and locks the chromophore against the acid- and heat-driven demetallation that would otherwise convert native chlorophyll to dull olive-brown pheophytin.

Common Markets Grades

Copper Chlorophyll is graded by total copper-pigment content and by free ionic copper level, with the latter strictly limited by food-additive monographs.

The oil-suspension grade carries 1 to 5 percent active copper chlorophyll dispersed in refined vegetable oil, with mixed tocopherols added for oxidative protection.

A 2 to 4 percent paste grade in a propylene-glycol or hardened-fat carrier serves confectionery and chewing-gum applications. Powdered grades at 0.5 to 2 percent on a maltodextrin or modified-starch carrier are used for dry blends, seasonings, and tableted supplements.

A concentrated pharmaceutical grade above 8 percent active is produced for ointment bases, deodorant tablets, and wound-care formulations where the deep green colour and stability are both required.

Regulatory Status

Copper Chlorophyll is approved as a food colour in the European Union under E141(i), covering the oil-soluble copper chlorophyll, and E141(ii), covering the sodium and potassium salts of the saponified water-soluble form.

The 2015 EFSA re-evaluation re-confirmed the additive in the Union list, with a maximum free ionic copper limit set by Commission Regulation specifications to ensure that the bound copper remains within the porphyrin ring.

In the United States, 21 CFR 73.125 approves the sodium copper chlorophyllin form for use in citrus-based dry beverage mixes. The oil-soluble copper chlorophyll itself does not have a separate FDA colour-additive listing for general food use, although it is widely used in dietary supplements and cosmetics.

The substance is listed in the Japanese Standards for Food Additives and in the Korean Food Additives Code. Halal, Kosher, and vegan certifications are routinely available when the carrier oil and plant feedstock are appropriately qualified.

Manufacturing Process

Copper Chlorophyll is produced from a native chlorophyll oleoresin extracted from alfalfa, nettle, or other green plant biomass with food-grade solvents.

The native magnesium-porphyrin complex is first converted to its acid-stable pheophytin form by mild acid treatment, which displaces the magnesium and leaves two protons in its place. The pheophytin is then reacted with a copper salt in a controlled aqueous-organic medium, where the copper inserts into the porphyrin pocket to give copper pheophytin, the regulated E141(i) substance.

The reaction is monitored spectrophotometrically by the shift of the red absorption maximum from near 667 nm in pheophytin to near 648 nm in the copper complex. Excess copper is removed by repeated washing and by chelation with food-grade sequestrants until the free copper level meets the monograph limit.

The purified copper complex is redissolved in refined vegetable oil or formulated into a propylene-glycol paste, deodorised under vacuum, and standardised against the copper-pigment assay before release.
