Pqq

Pqq, the abbreviated industry name for Pyrroloquinoline Quinone, is a redox-active tricyclic ortho-quinone (C₁₄H₆N₂O₈, molar mass 330.21 g/mol, CAS 72909-34-3). The molecule appears as a deep red-brown crystalline powder that is sparingly soluble in water and acidic media.

It serves as a redox cofactor in bacterial dehydrogenases and supports mitochondrial biogenesis in mammalian cells. Its catalytic turnover number, several orders of magnitude higher than typical quinones, is a defining functional feature.

Regulatory Status

Pqq is a permitted dietary ingredient in the United States, with the disodium salt holding New Dietary Ingredient notification status as BioPQQ. The free acid form is generally sold for research and pharmaceutical use rather than oral supplementation.

In the European Union, the disodium salt was authorised as a novel food in 2018 for use in food supplements within defined intake limits. The free acid itself has not been independently authorised for general food use.

Japan recognises the salt form under its Foods with Function Claims framework. Kosher and Halal certifications exist from major fermentation suppliers.

The substance carries no controlled-substance scheduling in major markets.

Industrial Pqq is produced by bacterial fermentation. Methylotrophic bacteria such as Hyphomicrobium denitrificans and selected pseudomonads naturally secrete the molecule when grown on methanol or related one-carbon substrates.

After fermentation, the broth is filtered, acidified, and the quinone is recovered by ion-exchange chromatography or selective precipitation. Subsequent crystallisation from acidic aqueous media yields the free acid in pharmacopoeial purity.

A total chemical synthesis exists, originally reported by Corey and Tramontano in 1981, but it cannot compete with fermentation on cost or yield at commercial scale.

Final material is dried under vacuum at low temperature and packaged under nitrogen to preserve the redox-active quinone moiety.
