Rebaudioside C

Rebaudioside C is a steviol glycoside isolated from the leaves of Stevia rebaudiana, distinguished from the other members of the family by carrying a rhamnose unit in its sugar chain alongside glucose. It is also recorded in older literature as dulcoside B.

The compound is the third most abundant glycoside in most stevia cultivars, typically accounting for several percent of dry leaf weight. Its sweetness intensity is lower than that of stevioside or rebaudioside A, in the region of 30 times sucrose, and its flavour profile is more strongly marked by liquorice and bitter notes. For that reason it is rarely used alone and is normally handled either as a component of total steviol glycoside preparations or as an isolated fraction for flavour and sweetness modulation work.

It comes as a white to off-white crystalline powder. Solubility in cold water is limited compared with rebaudioside A, and improves markedly with temperature and in hydroalcoholic systems. The glycoside is stable across the pH range of carbonated and still beverages, tolerates pasteurisation and retort conditions, and does not brown or caramelise during baking.

Isolated rebaudioside C is traded on assay, with material commonly offered at 80 percent and 95 percent minimum by HPLC, and the balance is other steviol glycosides. A far larger volume moves as total steviol glycoside powder in which rebaudioside C is one of the declared constituents rather than the traded specification.

Particle size is offered as standard powder and as milled fine powder for dry blending. Spray-dried grades carried on maltodextrin or blended to a defined sucrose equivalence are also produced where a formulator needs a directly dosable material rather than a pure glycoside.

Regulatory Status

In the United States, steviol glycosides purified to not less than 95 percent total glycosides are the subject of GRAS notices to the Food and Drug Administration for use as a general purpose sweetener, and rebaudioside C is one of the named glycosides covered by that purity basis. Isolated material below that purity threshold is not covered by the same notices.

In the European Union, steviol glycosides obtained from stevia leaf are authorised under E 960a within the food additives framework, with rebaudioside C listed among the specified glycosides. JECFA has evaluated steviol glycosides and maintains a specification monograph covering the named glycosides, and a corresponding Codex specification exists for the group.

Manufacturing Process

Dried stevia leaf is extracted with hot water or dilute ethanol to bring the glycoside fraction into solution. The crude extract is clarified to remove leaf solids and colloidal material, then treated with lime or a flocculant to precipitate pigments, proteins and tannins.

The clarified liquor passes over macroporous adsorbent resin, which binds the glycosides while sugars and salts pass through. The resin is eluted with ethanol and the eluate concentrated. Separation of rebaudioside C from stevioside and rebaudioside A relies on preparative or simulated moving bed chromatography, since the glycosides differ only in their sugar substitution and cannot be split by simple crystallisation. The enriched fraction is crystallised from aqueous ethanol, recrystallised where a higher assay is required, then centrifuged, dried and milled.
