Siberian Ginseng Extract

Siberian ginseng extract is prepared from the root, rhizome and sometimes the stem of Eleutherococcus senticosus, a thorny shrub of the Araliaceae found in far eastern Russia, north-eastern China, Korea and Japan. Despite the trade name it is not a Panax species and contains no ginsenosides, and it is increasingly labelled eleuthero to avoid that confusion.

The marker constituents are the eleutherosides, a chemically mixed group rather than a single class. Eleutheroside B is the phenylpropanoid syringin, eleutheroside E is the lignan syringaresinol diglucoside, and the group also takes in coumarins, triterpene saponins and polysaccharides. Because the eleutherosides are structurally unrelated to one another, a total eleutheroside figure only means something when the method and the markers used are stated.

The commercial article is a light to dark brown hygroscopic powder with a faintly bitter, woody taste. Standardised extracts made with aqueous alcohol dissolve or disperse in water and in dilute ethanol, and the glycosidic markers are reasonably stable to the temperatures of tabletting and spray drying but degrade under prolonged heat in acid. Liquid and fluid extract forms are also traded.

Common Markets Grades

Grades are set by marker content and by the plant part. Extracts standardised to 0.8 or 1.0 percent eleutherosides B and E measured by HPLC are the reference article, with 0.3 and 0.5 percent grades sold at lower cost and higher-strength material available. Ratio-only descriptions such as 10 to 1 or 20 to 1 are quoted throughout the trade but carry no assurance of marker content and should be read alongside an assay.

Root and rhizome material is the traditional and better-documented raw material, while stem and leaf extracts are cheaper and differ in composition, so buyers who need pharmacopoeial conformity should specify root and rhizome explicitly. Water extracts, aqueous ethanol extracts and polysaccharide-enriched grades are all produced, as are milled raw root powder and liquid fluid extracts at defined drug-to-extract ratios. Adulteration with related Acanthopanax species and with Periploca sepium has been documented, so an identity test rather than a marker assay alone is worth specifying.

Regulatory Status

In the United States eleuthero is used as a dietary ingredient in supplements with a documented history of use, and root extracts are also used in beverages, where suppliers rely on GRAS conclusions for the intended use rather than on a food additive listing. Labelling practice has moved away from the name Siberian ginseng, because United States labelling rules require that a product labelled ginseng be from a Panax species, so material from Eleutherococcus senticosus should be declared as eleuthero. Claims are structure and function claims with the standard disclaimer.

In the European Union Eleutherococcus senticosus root has a monograph within the traditional herbal medicinal product framework, and the herbal medicine route rather than the food supplement route is the better established path for it in several member states. Where it is used in food supplements, national rules on botanicals vary. There is no E number, since the extract is not an additive, and no authorised health claim. Codex has not adopted specifications for the species, so the general Codex provisions on contaminants and hygiene for dried plant material apply.

Manufacturing Process

Dried root and rhizome is cleaned of soil and foreign roots, inspected for identity, and milled to a coarse grade that lets solvent penetrate without producing fines that block the filter. Extraction is carried out with water or aqueous ethanol, typically 50 to 70 percent, in a countercurrent or multi-stage batch system at moderate temperature, since the glycosidic markers hydrolyse if the extraction is run hot for long. Two or three extraction passes are usual, and the miscella from each is combined.

The combined extract is filtered and the alcohol is recovered under vacuum for reuse. The aqueous concentrate is clarified, often by holding cold to precipitate resins and waxes, and then either concentrated directly for a low-strength extract or passed over macroporous adsorption resin to enrich the eleutherosides and strip sugars and salts, which is the step that sets the assay. The purified concentrate is standardised against an HPLC assay, blended with maltodextrin or with plant-derived carrier where needed, and spray dried at a controlled outlet temperature. The powder is sieved, tested and packed in moisture barrier bags.
