Regulatory Status

In the United States, a protein hydrolysate made from an edible substrate using an enzyme cleared for food processing is treated as a food ingredient rather than as a food additive, and freeze drying is a physical operation that does not change that classification. Trypsin of animal pancreatic origin is among the enzyme preparations with established food processing use, and its status must be confirmed for the specific preparation employed. Products sold into the supplement trade fall under the dietary supplement framework, and any allergenic substrate, such as milk, soy or egg, must be declared.

In the European Union, food enzymes including trypsin are governed by the food enzyme regulation and the Union list process, and enzymes of animal origin must additionally comply with the hygiene rules for products of animal origin. Manufacturing Process

The chosen protein substrate is dispersed in water and given a controlled heat step that denatures the native structure so that the cleavage sites are exposed to the enzyme. The slurry is cooled to the working window of trypsin, generally 37 to 50 degrees Celsius, and adjusted to slightly alkaline pH, since trypsin loses activity below neutral pH. Food grade trypsin, most often of porcine pancreatic origin or a microbial trypsin-like protease, is dosed at a set enzyme to substrate ratio and the digest is held for several hours with continuous alkali addition to hold pH as cleavage releases acid groups.

Hydrolysis is stopped by a short heat treatment or by acidification that inactivates the enzyme, and the liquor is centrifuged and microfiltered to strip undigested residue and any fat. Ultrafiltration sets the molecular weight cut-off, and activated carbon or nanofiltration removes colour, salts and bitter fractions where required. The permeate is concentrated under vacuum at low temperature, spread in trays and frozen, then loaded into a lyophiliser where primary drying sublimes the ice under vacuum at low shelf temperature and secondary drying removes bound water. The cake is broken out, milled and sieved in a humidity controlled room and packed immediately into moisture barrier containers.
