Vitamin K2

Vitamin K2 is the bacterial form of the fat-soluble vitamin K family, sold as menaquinone-7 (MK-7) or menaquinone-4 (MK-4).

MK-7 carries the CAS number 11032-49-8 and MK-4 carries 863-61-6. The two forms share the 2-methyl-1,4-naphthoquinone head common to all vitamin K compounds, but differ in the length and saturation of the isoprenoid side chain attached at C3.

Menaquinones reach the market in liquid and dry formats at activity-keyed potencies.

MK-7 from natto fermentation runs at 0.2 to 1.5 percent assay in medium-chain triglyceride oil, with all-trans isomer content specified separately because only the all-trans form is biologically active. Crystalline MK-7 at 96 percent assay serves high-potency supplement and pharmaceutical formulation.

Synthetic MK-7 produced by chemical assembly is also available and delivers a defined all-trans content, with batches typically certified above 99 percent geometric purity.

MK-4 is a yellow crystalline powder at 96 to 98 percent assay, with USP and Ph. Eur. monograph compliance. Cold-water-dispersible 1 percent powder and microencapsulated grades on modified starch carriers serve tablet, sachet, and beverage applications. Soft-gel oil dilutions at 1000 to 2000 µg per gram are the dominant supplement format.

Regulatory Status

Vitamin K2 menaquinones are authorised for use in food supplements and fortified foods in most regulated markets.

The European Union added menaquinone (MK-7) from Bacillus subtilis natto to the food-supplement vitamin-source list in 2009 after a positive EFSA opinion. Synthetic all-trans MK-7 was authorised as a novel food in 2014. The broader vitamin K source provisions of Directive 2002/46/EC permit MK-4.

The U.S. Food and Drug Administration recognises menaquinone-7 from natto as GRAS through Generally Recognised As Safe notifications. The Daily Value for vitamin K is 120 µg, expressed as phylloquinone equivalents.

Japan has the longest regulatory history with MK-4, where it is approved as a prescription drug for osteoporosis at high dose, and with MK-7 from natto, which has been consumed as a traditional food for centuries.

Pharmacopoeial monographs for menatetrenone (MK-4) appear in the Japanese Pharmacopoeia. Halal and Kosher certifications are commonly available, with vegan certification depending on the fermentation substrate and downstream processing aids.

Two production routes account for most global supply of vitamin K2.

MK-7 is predominantly produced by fermentation using Bacillus subtilis subsp. natto, the same organism that produces the traditional Japanese fermented soyfood natto. Submerged or solid-state fermentation on a glycerol or soy substrate yields a biomass rich in MK-7. The lipid fraction is recovered by solvent extraction, concentrated by molecular distillation, and crystallised to upgrade the all-trans isomer.

A chemical route to MK-7 uses menadione and a synthetic heptaprenyl side chain assembled by sequential coupling of isoprene units, with care taken to preserve the all-trans geometry across all six double bonds in the side chain. The chemical route delivers higher and more consistent all-trans content but does not qualify for natural-source labelling.

MK-4 is produced almost exclusively by chemical synthesis from menadione and geranylgeraniol. The geranylgeraniol side chain has four double bonds, all of which are required in the trans configuration for biological activity. The principal manufacturers operate in Japan, Norway, the United States, and China.
