D-Mannose

D-Mannose is a six-carbon aldohexose sugar and a C-2 epimer of glucose.

The CAS number is 3458-28-4 and the molecular formula is C₆H₁₂O₆ with a molar mass of 180.16 g/mol. The crystalline powder is white, freely water-soluble, and slightly less sweet than glucose.

D-Mannose is best known as a urinary tract health supplement. It is absorbed slowly from the gut, excreted largely unchanged in urine, and competes with bacterial adhesion proteins for binding sites on bladder epithelium, which is the mechanistic basis for its use in recurrent urinary tract infection prevention.

Supplement grade declares 98 to 99 percent purity and is a free-flowing white crystalline powder, often pre-sized for stick pack and capsule manufacture.

Pharmaceutical grade meets tighter limits on heavy metals and microbial counts and is used in registered medicinal products in some jurisdictions.

Glycobiology research grade runs at 99.5 percent or higher purity for use as a substrate or building block in glycoconjugate synthesis.

Regulatory Status

D-Mannose is recognised as a food and dietary supplement ingredient in most major markets without an E number.

The U.S. FDA accepts D-mannose as a dietary supplement ingredient under DSHEA. It is permitted in foods and supplements in the European Union as a normal carbohydrate ingredient.

In some European countries D-mannose is registered as a medical device or pharmaceutical product specifically for urinary tract health.

Manufacturing Process

D-Mannose is produced from plant-source mannan-rich polysaccharides or from glucose by enzymatic isomerisation.

The plant route hydrolyses mannan-rich biomass such as ivory nut, konjac glucomannan, or coffee residue under mild acid or with mannanase enzymes to release free mannose. The hydrolysate is purified by carbon, ion exchange, and chromatographic separation from accompanying glucose.

The biotech route starts from glucose and uses cellobiose 2-epimerase or related enzymes to epimerize glucose to mannose. The reaction is followed by chromatographic separation of mannose from unreacted glucose.

The purified mannose solution is concentrated and crystallised. The crystals are washed, dried, and milled.
