Fructooligosaccharides

Fructooligosaccharides are the plural form of the prebiotic carbohydrate class commonly abbreviated FOS, covering a distribution of glucose-terminated short-chain fructans with two to nine fructose units linked by β-2,1 bonds.

The plural label is the form commonly used on ingredient declarations and in clinical literature, because commercial preparations are always mixtures of GF2, GF3, and GF4 species rather than a single pure compound.

The material is partly sweet, freely soluble, non-digestible by human small intestine enzymes, and selectively fermented by bifidobacteria and lactobacilli in the colon.

Common Markets Grades

Fructooligosaccharides are graded by purity, source, and physical form.

Standard commercial grade declares 95 percent FOS on a dry basis, sold as either spray-dried powder or 75 percent solids syrup.

Chromatographically separated grade declares 97 to 99 percent FOS for high-purity supplement and infant nutrition use.

Inulin-derived oligofructose grade is produced from chicory and carries a slightly wider DP distribution (DP 2 to 7) than sucrose-source FOS.

Organic and non-GMO certified grades exist from both chicory and sucrose sources.

Regulatory Status

Fructooligosaccharides are recognised as a food ingredient and dietary fibre in most major markets, generally without an E number.

The U.S. FDA accepts FOS as a self-affirmed GRAS ingredient and lists short-chain FOS as a dietary fibre under the revised Nutrition Facts panel.

EFSA has reviewed FOS for infant nutrition. Japan recognises FOS under the Foods for Specified Health Use framework.

The Codex Alimentarius covers FOS within the general dietary fibre definition based on the AOAC analytical methods.

Manufacturing Process

Commercial fructooligosaccharides are produced by two parallel routes.

The sucrose-substrate route uses immobilized fructosyltransferase enzymes (commonly from Aspergillus or Aureobasidium) that transfer fructose units between sucrose molecules to build short-chain fructans.

The chicory-inulin route uses endo-inulinase to partially hydrolyse the long-chain inulin from chicory root into shorter oligofructose fragments.

Both reaction streams are purified through carbon treatment and ion exchange. Chromatographic polishing separates residual mono- and disaccharides for high-purity grades.

The final solutions are concentrated and dried into white free-flowing powders or kept as clear syrups for liquid dosing.
