Echinacoside

Echinacoside is a phenylethanoid glycoside (PhG) and the principal active marker of Cistanche tubulosa and Cistanche deserticola, two parasitic desert herbs traditionally used in Chinese tonic medicine.

The molecule (CAS 82854-37-3, formula C₃₅H₄₆O₂₀, molar mass 786.7 g/mol) is a caffeoyl-glycoside built on a hydroxytyrosol aglycone, decorated with glucose, rhamnose, and a second caffeic-acid ester. It presents as a light-yellow amorphous powder, freely soluble in water and methanol.

Regulatory Status

Cistanche stem and its extracts have a long history of use in China and are listed in the Chinese Pharmacopoeia.

In the United States, Cistanche tubulosa stem extract is marketed as a dietary ingredient under DSHEA, with several New Dietary Ingredient notifications on file covering specified phenylethanoid glycoside content.

In the European Union, Cistanche tubulosa stem extract is the subject of Novel Food assessment, with status that depends on extraction method, daily dose, and intended food category.

In Japan, Cistanche has a long history of use as a traditional tonic ingredient and is treated in commerce as a non-medicinal food material.

Echinacoside itself is not approved as a drug substance, and disease-treatment claims are not permitted on supplement labels.

Manufacturing Process

Commercial echinacoside is produced almost exclusively from the fleshy stems of Cistanche tubulosa, harvested in the Taklamakan and Gobi desert regions of Xinjiang and Inner Mongolia, with smaller volumes from Cistanche deserticola.

The harvested stems are sliced, sun-dried, and milled. Extraction is carried out with hot water or dilute ethanol, with the crude extract concentrated under vacuum.

Lower-strength grades (10 to 30 percent) are produced by adsorbing the extract onto macroporous resin (typically D101 or AB-8), washing off polar sugars and salts with water, then desorbing the phenylethanoid fraction with 30 to 60 percent ethanol. The eluate is concentrated and spray-dried.

High-purity grades (50 percent and above) require a second resin pass or preparative reversed-phase chromatography, often with a final recrystallisation or freeze-drying step.

Sustainable cultivation of Cistanche tubulosa on cultivated Tamarix host trees has expanded sharply in the past decade, reducing pressure on wild-harvested material.
