Isomaltulose

Isomaltulose is a disaccharide of glucose and fructose linked α-1,6, in contrast to the α-1,2 linkage of sucrose.

The CAS number is 13718-94-0 and the molecular formula is C₁₂H₂₂O₁₁. It is marketed under the trade name Palatinose by Beneo (formerly Südzucker).

The shifted glycosidic bond is hydrolysed by intestinal isomaltase much more slowly than sucrose is hydrolysed by sucrase, which gives isomaltulose a low glycemic index and sustained energy release while still delivering the full caloric value of sucrose.

Isomaltulose is produced mainly as the monohydrate crystal in food and beverage grade.

The standard grade is a free-flowing white crystal with sweet, sucrose-like taste at about half the intensity of sucrose.

Powdered and fine-particle grades are produced for direct-compression confectionery and tablet applications.

A liquid syrup grade is produced for inline beverage and dairy dosing.

Regulatory Status

Isomaltulose is approved as a novel food in the European Union since 2005 and is permitted in foods generally, with a labelling requirement that the product is more slowly digested than sucrose.

The U.S. FDA granted GRAS status under GRN 000184. Australia, New Zealand, China, Japan, and most Latin American and Asian markets have parallel approvals.

JECFA reviewed isomaltulose and concluded no specified ADI was required.

The European tooth-friendly health claim under Regulation 432/2012 applies to isomaltulose in place of sucrose.

Manufacturing Process

Isomaltulose is produced from sucrose by enzymatic isomerisation.

A sucrose solution is passed through a column of immobilized cells of Protaminobacter rubrum or a related organism that expresses sucrose isomerase. The enzyme cleaves the α-1,2 bond of sucrose and re-forms it as an α-1,6 bond, yielding isomaltulose.

The conversion proceeds with high selectivity and the reaction mass contains around 80 percent isomaltulose along with smaller amounts of trehalulose and unreacted sucrose.

The solution is purified through ion-exchange and carbon treatment, concentrated, and crystallised as the monohydrate.
