Oxyresveratrol

Oxyresveratrol is a hydroxylated stilbene, chemically 2,3',4,5'-tetrahydroxy-trans-stilbene, with molecular formula C14H12O4 and molar mass 244.24 g/mol. It is the trans-stilbene skeleton of resveratrol carrying one additional hydroxyl group, and that extra phenolic function is responsible for its markedly stronger antioxidant behaviour and its potency as a tyrosinase inhibitor.

It occurs naturally in the heartwood of Artocarpus lakoocha, in mulberry wood and root bark of Morus alba, and in Smilax and Schoenocaulon species. The heartwood of Artocarpus lakoocha, sold in Southeast Asia as the traditional preparation puag-haad, is the richest known source and is the usual industrial feedstock.

Commercial material is an off white to pale beige crystalline powder, practically insoluble in water, freely soluble in ethanol, methanol and dimethyl sulphoxide, and soluble in propylene glycol and glycerine blends used for topical formulation. Like other stilbenes it isomerises from the trans to the less active cis form under ultraviolet light and darkens at alkaline pH, so it is handled away from light and formulated on the acid side.

Common Markets Grades

Trade covers two distinct levels. Standardised plant extracts of Artocarpus lakoocha heartwood or mulberry are sold at 5, 10 and 20 percent oxyresveratrol by HPLC and retain the accompanying stilbene and flavonoid fraction of the wood. Purified grades are sold at 90, 95 and 98 percent as a single isolated compound, with the trans isomer content stated separately from total assay because the cis form is inactive for the tyrosinase application.

Physical options are limited to crystalline powder of defined particle size, usually passing 80 or 100 mesh, and to pre-dispersed liquid concentrates in propylene glycol, butylene glycol or ethanol for cosmetic use, typically at 1 to 5 percent active. Micronised and cyclodextrin complexed versions are made where dissolution rate in an aqueous base matters.

Regulatory Status

Oxyresveratrol is used principally as a cosmetic ingredient rather than as a food additive. In the United States it has no food additive regulation and no listing among substances generally recognised as safe, so food use would require a GRAS conclusion or a food additive petition that has not been established for this compound. Cosmetic use falls under the general provisions of the Federal Food, Drug, and Cosmetic Act, which places the safety obligation on the manufacturer without pre-market approval, and skin lightening claims that cross into treatment of a disease would move a product into the drug category.

In the European Union oxyresveratrol has no E number and is not an authorised food additive. As a purified stilbene it would fall under Regulation 2015/2283 on novel foods, and no authorisation covering it has been granted, so it is not marketable as a food or food supplement ingredient in the Union. Regulation 1223/2009 governs cosmetic use, and the substance appears in neither the prohibited nor the restricted entries, so it rests on a safety assessment in the product information file. Codex and JECFA have not evaluated it.

Manufacturing Process

Dried heartwood of Artocarpus lakoocha, or mulberry root bark and wood chips, is milled to a coarse powder and extracted with aqueous ethanol at 60 to 80 degrees Celsius in a counter current or multiple stage percolation battery. The crude extract is filtered, the ethanol recovered under vacuum, and the aqueous concentrate held cold so that a large part of the stilbene fraction crystallises directly, since oxyresveratrol is poorly water soluble. This crude crystal crop is the basis of the standardised extract grades.

For purified material the crude solid is redissolved in hot ethanol or ethyl acetate, treated with activated carbon to remove wood pigments and tannins, and passed over a macroporous adsorption resin or a preparative chromatography column to separate oxyresveratrol from resveratrol and the accompanying stilbene glycosides. The purified fraction is concentrated, recrystallised from an alcohol and water mixture, filtered on a centrifuge and dried under vacuum at low temperature and protected from light. The dried crystals are milled, assayed by HPLC for total and trans isomer content, and packed in light resistant containers under nitrogen.
