Common Markets Grades
Black pepper extract is graded by piperine content, measured by HPLC at 343 nm.
Mainstream supplement grades are 95, 97, and 98 percent piperine. The 95 percent grade is the global benchmark and the specification used in most clinical-trial work on bioavailability enhancement.
Higher grades at 99 percent piperine are produced for pharmaceutical-style formulations and for branded ingredients that require tighter assay limits.
Lower-grade pepper extracts at 50 and 85 percent piperine are made for food and spice applications where pungency rather than bioavailability is the objective.
Whole black pepper oleoresins at 30 to 50 percent piperine are sold to the savory-flavour and seasoning category.
Material is a fine off-white to pale-yellow crystalline powder with a sharp pungent note. Microencapsulated and beadlet versions exist for taste-masked supplement applications.

Regulatory Status
Black pepper and its extracts have universal acceptance as food ingredients and traditional spices.
In the United States black pepper extract and piperine are GRAS for use as flavour ingredients and are sold as dietary-supplement ingredients under DSHEA. At least one branded high-purity piperine preparation carries an FDA GRAS notification.
In the European Union black pepper extract is a recognised food ingredient. Standardised piperine preparations entering the food-supplement channel are evaluated at the member-state level.
In Japan piperine is recognised as a food flavouring and supplement ingredient.
The European Food Safety Authority has evaluated piperine and identified it as a CYP3A4 and P-glycoprotein inhibitor, with relevant drug-interaction caveats for high-dose supplement use.
Manufacturing Process
Black pepper is sourced from cultivated Piper nigrum vines in India (Kerala and Karnataka), Vietnam, Indonesia, Sri Lanka, and Brazil. Vietnam is the largest global producer of pepper berries, while India leads in piperine extraction capacity.
Unripe green berries are harvested, briefly blanched, and sun-dried for several days. The fruit skin oxidises during drying to produce the characteristic wrinkled black peppercorn.
Dried peppercorns are cleaned, milled, and extracted with a polar organic solvent such as ethanol, methanol, or dichloromethane. The choice of solvent and extraction temperature drives the selectivity for piperine over the essential-oil fraction.
The crude extract is concentrated, washed to remove volatile oils, and crystallised from ethanol. Multiple recrystallisation cycles bring the assay from a crude 40 to 60 percent up to the 95 to 99 percent commercial grades.
For the higher grades the crystalline piperine is washed with cold ethanol, recrystallised, and dried under vacuum at low temperature to preserve crystal structure and minimise residual solvents.
HPLC with UV detection at 343 nm against a piperine reference is the standard assay. Residual solvent and heavy-metal testing complete the certificate of analysis.
