Trehalose
Trehalose is a non-reducing disaccharide composed of two D-glucose units linked α,α-1,1 through their anomeric carbons.
The CAS number is 99-20-7 and the molecular formula is C₁₂H₂₂O₁₁. The unique α,α-1,1 linkage makes the molecule unusually stable to heat, acid, and the Maillard reaction.
Trehalose is about 45 percent as sweet as sucrose, dissolves cleanly without aftertaste, and acts as a powerful cryoprotectant and protein stabiliser in food, pharmaceutical, and biotechnology applications.

Food, pharmaceutical and high-purity research grades of trehalose all use the dihydrate crystal.
Food grade meets the JECFA and Codex specifications and comes as a free-flowing white crystal or fine powder.
Pharmaceutical grade meets USP-NF, Ph. Eur., and JP monographs and is used in lyophilized injectable formulations and oral solid dose.
Cell-culture and bioprocess grade carries tight endotoxin limits and low residual metal content for biopharmaceutical use.
An anhydrous trehalose grade is produced for moisture-sensitive applications, although the dihydrate is the standard form.

Regulatory Status
Trehalose is approved as a food ingredient in the European Union, United States, Japan, China, and most other major markets.
The U.S. FDA granted GRAS status under GRN 000045 in 2000. The EU authorised it as a novel food in 2001.
JECFA reviewed trehalose and concluded no specified ADI was required on safety grounds.
Pharmacopoeial monographs appear in the USP-NF, European Pharmacopoeia, and Japanese Pharmacopoeia.
Manufacturing Process
Modern trehalose production uses an enzymatic process on starch developed by Hayashibara of Japan in the 1990s.
Liquefied starch is treated sequentially with maltooligosyl trehalose synthase (MTSase) and maltooligosyl trehalose trehalohydrolase (MTHase). MTSase converts the terminal maltooligosaccharide α-1,4 linkage to the α,α-1,1 trehalose linkage, and MTHase cleaves the trehalose unit from the remaining chain.
The reaction mass is treated with glucoamylase to convert residual oligosaccharides to glucose, which is then separated by chromatography.
The purified trehalose solution is decolorised, demineralized, concentrated, and crystallised as the dihydrate.
