Copper Gluconate

Copper Gluconate is the copper salt of D-gluconic acid, with the empirical formula Cu(C6H11O7)2 and CAS number 527-09-3. It contains approximately 14 percent elemental copper.

The compound is a fine light blue powder that dissolves freely in water to give a clear blue solution. Its gentle taste, low gastric irritation, and broad food regulatory clearance have made it the default copper carrier in human supplements, food fortification, and infant nutrition.

USP grade is graded at 97.0 to 102.0 percent assay on the dried basis, with ICH Q3D compliant limits on lead, arsenic, cadmium, and mercury. This grade serves solid oral supplements, liquid drops, and infant nutrition.

FCC food grade meets the same purity profile and is produced with controls on reducing substances and chloride. It is the dominant grade for fortified beverages, cereals, and nutrition bars.

The U.S. Food and Drug Administration recognises Copper Gluconate as GRAS for use as a nutrient under 21 CFR 184.1260. It is also covered by a United States Pharmacopeia monograph for use in dietary supplements and pharmaceutical preparations.

The European Union authorises Copper Gluconate as a source of copper for food supplements under Annex II of Directive 2002/46/EC, for fortified foods under Regulation (EC) No 1925/2006, and for infant formula and follow-on formula under Regulation (EU) 2016/127.

JECFA has evaluated copper gluconate within its broader assessment of copper compounds and considers it acceptable as a nutrient source for food and supplement use.

Pharmacopoeial monographs appear in the United States Pharmacopeia (USP) and the European Pharmacopoeia (Ph. Eur.). Halal and Kosher certifications are routinely available, as is non-GMO documentation for fermentation-derived gluconic acid.

Manufacturing Process

Copper Gluconate is produced by acid-base reaction of D-gluconic acid with a copper source.

Gluconic acid, manufactured by the aerobic fermentation of glucose with Aspergillus niger, is filtered, decolorised, and concentrated. It arrives as a 50 percent aqueous solution or as glucono-delta-lactone for tighter water control.

The gluconic acid solution is diluted to 20 to 25 percent and warmed to 60 to 80 degrees Celsius. High-purity copper carbonate (basic) or freshly precipitated copper hydroxide is added at the stoichiometric two-to-one molar ratio (gluconate to copper). Carbon dioxide is liberated when copper carbonate is used, supporting end-point control.

The clear blue solution is treated with activated carbon to remove fermentation pigments, polish-filtered, and concentrated under vacuum. Crystallisation is induced by cooling or by ethanol antisolvent precipitation.

The wet cake is washed, dried under vacuum at 40 to 60 degrees Celsius, milled, and packed under low-humidity conditions. Each lot is tested by ICP-MS for heavy metals and by titration for assay.
