Glucosyl Steviol Glycosides

Glucosyl Steviol Glycosides are steviol glycosides that have been enzymatically modified by adding alpha-linked glucose units onto the native glycoside sugar chains.

The modification is carried out with cyclodextrin glucanotransferase (CGTase) in the presence of a starch-derived glucose donor. The result is a broad mixture of glucosylated stevioside, glucosylated Reb A, and minor species, with improved solubility and a softer sweet profile compared with the starting material.

The product is generally specified by total glucosyl steviol glycoside content and by the ratio of mono-, di-, tri-, and tetra-glucosyl species.

Standard food-grade material runs at 80 to 95 percent total glycosides on a dry basis, with the remainder consisting of residual reaction sugars.

High-purity grades at 95 percent and above are produced for clear beverage applications where a fully soluble, neutral-colour powder is required.

Blended grades combine glucosyl steviol glycosides with parent stevia extract or with Reb A to fine-tune sweetness onset and persistence at a defined cost point.

Regulatory Status

Glucosyl Steviol Glycosides are recognised as a steviol glycoside sub-category in major regulated markets.

In the European Union the material is covered under E960c, the sub-listing introduced for enzyme-modified steviol glycosides. The U.S. Food and Drug Administration accepts the product under GRAS notices that cover the CGTase modification process.

JECFA has evaluated enzyme-modified steviol glycosides and concluded that the product meets the steviol glycosides specification, with the ADI expressed as steviol.

Kosher and Halal certifications are routinely available. Non-GMO Project verification is generally not available because the CGTase enzyme is often produced in genetically modified host organisms.

Manufacturing Process

Production starts with a purified steviol glycoside feed, typically stevia extract at 80 to 95 percent TSG, and a starch-derived glucose donor such as dextrin or partially hydrolysed starch.

The feed is dissolved in water and reacted with cyclodextrin glucanotransferase (CGTase). The enzyme transfers alpha-1,4 and alpha-1,6 linked glucose units from the donor onto the existing glucose residues of the steviol glycoside, generating a ladder of glucosylated species.

Reaction control over enzyme dose, donor-to-acceptor ratio, time, and temperature sets the average chain length. Longer chains increase solubility and soften sweetness, but reduce per-weight potency.

The reaction mass is heat-treated to inactivate the enzyme, decolorised over activated carbon, deionized, and concentrated. The product is spray dried to a free-flowing white powder.
