B. longum subsp. longum is one of the three subspecies of Bifidobacterium longum and is a dominant member of the adult colonic microbiota, a Gram positive, non spore forming, strictly anaerobic rod that ferments carbohydrate by the fructose-6-phosphate phosphoketolase pathway to acetate and lactate.
The trade material is a cream to light beige free flowing powder with a faint fermented odour. Its strength is declared as colony forming units per gram rather than as a chemical assay, and the fermentation biomass is diluted with maltodextrin or microcrystalline cellulose to the potency the finished dose form requires.
Because the organism is a strict anaerobe, oxygen exposure limits shelf life as much as moisture and temperature do. The powder is hygroscopic, is dosed under low humidity, and is packed under nitrogen in aluminium foil laminate and stored refrigerated or frozen. It disperses in cold water but cannot be carried through pasteurisation, extrusion or spray drying, so it is added after every thermal step in a process.

In the United States Bifidobacterium longum is used in conventional foods on a GRAS basis, with notified GRAS conclusions covering specific strains including use in infant formula, and is marketed as a dietary ingredient under the dietary supplement framework. Digestive or immune statements are structure and function claims subject to supplement labelling rules rather than authorised health claims, and the main label obligations are the strain designation and the declared viable count.
In the European Union live cultures of this kind are food ingredients rather than additives, so no E number applies. Bifidobacterium longum holds qualified presumption of safety status at species level in the EFSA framework, while most member states restrict the term probiotic on labels because no health claim for these cultures carries authorisation. The joint FAO and WHO guidelines on the evaluation of probiotics in food set the internationally recognised requirements for strain identification, viability declaration and labelling, and Codex has no strain specific standard for the organism.
Manufacturing Process
The strain is grown by submerged anaerobic batch fermentation. A working seed vial is expanded through flask and seed fermenter stages into a production vessel charged with a fermentable carbohydrate, a nitrogen source such as yeast extract or a dairy free peptone, mineral salts and a reducing agent to hold the redox potential low enough for a strict anaerobe. The headspace is blanketed with nitrogen or carbon dioxide throughout, and pH is held near neutral by automatic alkali dosing, since the acetate and lactate the culture produces would otherwise stop growth well before the target cell density is reached.
At harvest the broth is concentrated by continuous centrifugation or by membrane filtration to a thick cell cream. Cryoprotectants, typically sucrose, trehalose and a protein or hydrocolloid, are blended in to stabilise the cell membranes through freezing. The cream is pelletised in liquid nitrogen or frozen on trays and freeze dried until residual moisture falls to the low single figures. The dried cake is milled, assayed for viable count and strain identity, blended with carrier to the declared potency and packed under nitrogen.
