Magnesium L-threonate is the magnesium salt of L-threonic acid, a four carbon sugar acid derived from L-ascorbic acid, and sells as a white to off-white powder for nutritional supplement use.
The molecule carries one magnesium ion for two threonate anions and has a molecular weight near 294, which puts the elemental magnesium content close to eight percent. That is low against inorganic salts such as magnesium oxide, so a given magnesium dose needs a comparatively large powder weight, and the material is almost always presented in capsules or multi capsule daily servings rather than in tablets.
The salt is freely soluble in water, giving a clear, near neutral solution with a faintly sour taste from the threonate ligand. It is markedly hygroscopic, picking up moisture from humid air and caking if left open, so it is handled in controlled humidity rooms and packed with desiccant. Regulatory Status
In the United States magnesium L-threonate is marketed as a dietary ingredient, supported by new dietary ingredient notification and by safety studies submitted by the original developer, and finished supplements are produced under 21 CFR Part 111. It is not an approved food additive and is not affirmed as generally recognised as safe for general food fortification, so its use is restricted to supplement formats rather than conventional foods and beverages.
In the European Union the salt is not among the magnesium sources permitted for food supplements under the annex to Directive 2002/46/EC, and it has no novel food authorisation, so it is not legally saleable as a supplement ingredient in the EU. Status differs by market elsewhere, with several Asian and Middle Eastern jurisdictions treating it as a permitted supplement ingredient subject to registration. No Codex or JECFA specification monograph exists for the salt.
Manufacturing Process
Manufacture begins with L-ascorbic acid, which is oxidised in aqueous alkaline conditions, commonly with hydrogen peroxide in the presence of a calcium or magnesium base, to cleave the ring and give L-threonic acid. The oxidation is run at controlled temperature and pH to limit over oxidation to oxalate and other short chain acids, and the crude liquor is treated with activated carbon to strip colour bodies formed during the reaction.
The threonate is then converted to the magnesium salt, either by direct neutralisation of purified L-threonic acid with magnesium hydroxide or magnesium carbonate, or by ion exchange from an intermediate calcium threonate. The neutral liquor is filtered, decolourised again if needed, and concentrated under vacuum, after which the product is recovered by spray drying or by antisolvent crystallisation into ethanol. Because the salt is hygroscopic, drying, milling and packing are carried out in dehumidified rooms.
