Magtein delivers approximately 8 percent elemental magnesium, the lowest of any common magnesium supplement form. The trade-off is intentional, since the threonate carrier is held to be the active vehicle for blood-brain barrier transit.

The standard grade is a free-flowing white-to-off-white powder meeting USP-style limits for heavy metals, residual solvents, and microbiological purity. Particle size is tuned for capsule and stick-pack filling rather than direct tablet compression.

Generic non-Magtein magnesium L-threonate has appeared from Indian and Chinese manufacturers in recent years, typically at lower price points but without the clinical-trial dossier or the brand recognition that supports finished-product premium pricing.

Regulatory Status

Magnesium L-Threonate has cleared the relevant safety reviews for dietary-supplement use.

In the United States, AIDP filed a New Dietary Ingredient notification with the FDA in 2010, and Magtein subsequently received self-affirmed GRAS status for use in food and beverage products.

The European authorisation picture is more limited. Magnesium L-Threonate is not currently listed in Annex II of Directive 2002/46/EC, which restricts its use as a magnesium source in EU food supplements. Sales channels in the EU therefore route through novel food review or non-supplement formats.

Patent protection through the Magtein portfolio remains a key regulatory and commercial gatekeeper, with infringement actions taken against unlicensed cognitive-positioned imitators.

Manufacturing Process

The manufacturing route begins with L-ascorbic acid (vitamin C) as the starting material.

Controlled oxidative cleavage of L-ascorbic acid, typically with hydrogen peroxide under alkaline conditions, opens the lactone ring and yields L-threonic acid alongside oxalic acid. The two acids are separated by ion-exchange chromatography or selective crystallisation.

Purified L-threonic acid is then neutralised with magnesium carbonate or magnesium hydroxide in aqueous solution. The reaction produces magnesium L-threonate in a 1 to 2 magnesium to threonate ratio with evolution of carbon dioxide or water.

The resulting solution is concentrated, spray-dried or freeze-dried, and milled to specification. Strict control over residual oxalate, peroxide, and starting-material impurities is required, as these can affect both safety and clinical performance.

AIDP holds the supply rights for Magtein through licensing arrangements that originated with the MIT research team led by Guosong Liu. The patent portfolio covers both the composition of matter and the cognitive use claims.
