Neohesperidin Dihydrochalcone

Neohesperidin Dihydrochalcone is a semi-synthetic, high-intensity sweetener derived from neohesperidin, a bitter flavanone glycoside abundant in the peel of bitter (Seville) oranges. Catalytic hydrogenation opens the flavanone C-ring to give the corresponding dihydrochalcone, which is intensely sweet rather than bitter.

The molecule has the formula C₂₈H₃₆O₁₅ and a molar mass of 612.58 g/mol. It is an off-white to pale yellow crystalline powder, sparingly soluble in cold water and freely soluble in hot water, ethanol, and dilute alkaline solutions. Commonly abbreviated NHDC, the compound delivers a sweetness intensity of 1,500 to 1,800 times that of sucrose at typical use levels.

The sweetness profile is slow-onset and long-lasting, with a characteristic licorice and cooling menthol-like aftertaste that becomes more pronounced at higher concentrations. The distinctive finish makes NHDC most useful as a flavour enhancer, masking agent, and co-sweetener rather than as a standalone primary sweetener.

Regulatory Status

Neohesperidin dihydrochalcone is approved as a food additive in the European Union and a number of other markets, with more limited approval status in the United States.

In the European Union it carries the E number E959 under Regulation (EC) No 1333/2008, both as a sweetener and as a flavour enhancer, with use levels defined per food category in Annex II.

The U.S. Food and Drug Administration does not list NHDC under the general-purpose intense-sweetener provisions. The substance has been the subject of GRAS notifications in the United States for use as a flavour and flavour enhancer in specific food categories.

JECFA has evaluated neohesperidin dihydrochalcone and assigned acceptable status as a sweetener and flavour enhancer. Health Canada and several Asian and Latin American regulators permit its use under defined conditions. Halal and Kosher certifications are commonly available because the manufacturing process uses no animal-derived inputs.

Manufacturing Process

Neohesperidin dihydrochalcone is produced from neohesperidin, a flavanone glycoside that occurs at percent-level concentrations in the immature peel of bitter (Seville) orange, Citrus aurantium.

Bitter orange peel is extracted with hot water or aqueous ethanol, and neohesperidin is recovered from the extract by precipitation, crystallisation, or column chromatography. An alternative starting point is naringin, which is first converted to neohesperidin by methylation in some industrial routes.

The purified neohesperidin is dissolved in dilute alkaline solution to open the flavanone C-ring to the corresponding chalcone, which is then catalytically hydrogenated over a palladium-on-carbon catalyst under hydrogen pressure. The hydrogenation reduces the α,β-unsaturated ketone of the chalcone to give the dihydrochalcone, in which the sweet taste is established.

The crude dihydrochalcone is neutralised, decolorised with activated carbon, and crystallised from water or aqueous ethanol. The dried crystals are milled to the target particle size, screened, and packaged.
