Calcium Gluconate

Calcium Gluconate is the calcium salt of gluconic acid, with the formula Ca(C₆H₁₁O₇)₂ (CAS 299-28-5). It delivers approximately 9 percent elemental calcium, the lowest of the common pharmaceutical calcium salts, but in exchange dissolves readily in water and is well tolerated in the gut.

The salt is the standard parenteral calcium source in hospital practice and is also used as an oral supplement, food fortificant, and topical antidote for hydrofluoric acid burns.

Calcium Gluconate is produced primarily as the monohydrate.

USP-grade powder is the dominant pharmaceutical specification, used for sterile injectable manufacture and oral solid dosage formulation. The injectable raw material undergoes additional endotoxin testing and is dissolved into 10 percent w/v sterile solutions for intravenous administration.

Food Chemicals Codex grade covers calcium fortification, acidity regulation, and firming applications. Granular grades support direct compression of tablets and effervescent calcium products.

Topical-formulation grade is produced for the manufacture of 2.5 percent gels used as a first-aid antidote for skin exposure to hydrofluoric acid.

In the European Union it is listed as food additive E578, permitted as an acidity regulator, sequestrant, and firming agent. It is also approved as a calcium source for food supplements and fortified foods.

The U.S. Food and Drug Administration affirms it as GRAS under 21 CFR 184.1199 for general food use. Pharmacopoeial monographs appear in USP-NF, Ph. Eur., BP, and JP, with separate monographs covering Calcium Gluconate Injection and Calcium Gluconate Tablets.

The World Health Organisation includes Calcium Gluconate on the WHO Model List of Essential Medicines for the treatment of hypocalcemia and as an antidote for fluoride and oxalate poisoning.

Manufacturing Process

Calcium Gluconate is manufactured by neutralisation of gluconic acid with a calcium source.

Industrial gluconic acid is produced by submerged aerobic fermentation of glucose using Aspergillus niger or, increasingly, Gluconobacter species. The microbial enzyme glucose oxidase converts glucose to gluconic acid with high selectivity and yield.

The fermentation broth is clarified, then neutralised with calcium carbonate or calcium hydroxide. The resulting Calcium Gluconate is crystallised from the cooled solution, recovered, washed, and dried as the monohydrate.

Pharmaceutical-grade material requires additional decolorisation with activated carbon, ion-exchange polishing, and recrystallisation to meet pharmacopoeial purity for parenteral use. Injectable solutions are sterile-filtered and terminally sterilised after fill.
