Zinc Gluconate

Zinc Gluconate is the zinc salt of D-gluconic acid, with the empirical formula Zn(C6H11O7)2 and CAS number 4468-02-4. It contains approximately 14.3 percent elemental zinc.

The compound is a white to off-white crystalline powder that dissolves readily in water. Its gentle taste and superior gastrointestinal tolerance versus inorganic zinc salts have made it the default zinc carrier in cold-relief lozenges and consumer multivitamins.

USP and Ph. Eur. monograph material runs 97.0 to 102.0 percent assay calculated on the dried basis. Particle size is typically classified into fine powder for direct compression and granular grade for blending in pre-mixes.

Food grade conforms to FCC and JECFA specifications and is produced at the same assay window with controls on lead, arsenic, and reducing substances.

Lozenge-specific grades exist in spray-dried or roller-compacted form to improve flow and minimise the astringent metallic mouthfeel during slow oral dissolution.

Regulatory Status

Zinc Gluconate carries broad regulatory approval as a nutrient source.

In the United States it is GRAS for use as a nutrient under 21 CFR 182.8988. The FDA OTC drug framework regulates the cold-lozenge category, with zinc gluconate as a permitted active.

The European Union authorises Zinc Gluconate as a source of zinc in food supplements under Annex II of Directive 2002/46/EC, in foods for general fortification under Regulation (EC) No 1925/2006, and in foods for specific groups under Regulation (EU) No 609/2013.

JECFA evaluated zinc gluconate under the general zinc compound assessment and considers it an acceptable nutrient source.

Pharmacopoeial monographs exist in the United States Pharmacopeia (USP), the European Pharmacopoeia (Ph. Eur.), and the British Pharmacopoeia (BP). Halal and Kosher certifications are widely available.

Manufacturing Process

Industrial production of Zinc Gluconate follows a straightforward acid-base reaction.

D-Gluconic acid, manufactured by the controlled aerobic fermentation of glucose with Aspergillus niger, is the primary raw material. The fermentation broth is filtered, decolorised, and concentrated into a gluconic acid solution or recovered as glucono-delta-lactone.

The gluconic acid solution is reacted with zinc oxide, zinc carbonate, or freshly precipitated zinc hydroxide. The reaction proceeds at 50 to 80 degrees Celsius with mild agitation until pH stabilises at neutral. Excess zinc source is filtered off.

The resulting Zinc Gluconate solution is treated with activated carbon, polish-filtered, and crystallised by cooling or by anti-solvent precipitation with ethanol. The wet cake is washed, dried under vacuum, and milled to the target particle size.

Lead-free zinc oxide is used throughout to meet pharmacopoeial heavy-metal limits.
